| Objective: To reserch the relationship between human papillomavirus (HPV) type16E6/E7DNA viral load and development of cervical lesions. Discuss the feasibility andclinical application value of high-risk human papilloma virus (HR-HPV) E6/E7DNAtranscripts in cervical cancer samples by using liquid-based Thin-prep cytology test (TCT).Methods:1)243cases of TCT abnomaly detection diagnosis (ASCUS115cases, LSIL54cases, HSIL60cases, squamous cell carcinoma13cases), and129case with normal TCTtest. Real-Time PCR assays was performed to detected HPV16E6/E7DNA in liquid-basedcervical cytology. Result:1) The HPV16E6/E7positive of ASCUS, LSIL, HSIL, SCCwere69%,84.6%,91%,100%,59.2%in control group, there was a significant differenceamong the different stage cervical lesions (P<0.01);2) The median load of HPV16E6/E7were16.77,57.95;67.87,93.76;331844,8165.47;29096,176256.41;5.15,5.15respectively,there was a significant difference among the different grade of cervical lesions (P<0.05);here was significant correlation in the E6/E7viral load and grade of cervicaldysplasia(R=0.654, P<0.01and R=0.593, P<0.01);4) HPV16E6load in Uygur and Hanfemales were9.05,18.31in chronic cervicitisã€36.275,162.545in CINâ… ã€124.52,337.37in CINâ…¡ã€CINâ…¢2003.64,1046.30ã€24373.12,22500.78in SCCï¼›HPV16E7load in Uygurand Han females were18.82,71.70in chronic cervicitisã€85.23,181.09in CINâ… ã€350.67,462.36in CINâ…¡ã€1338.95,3419.35in CINâ…¢ã€48115.94,255705.83in SCC。HPV16E6/E7load high in Uygur compare with Han females (P<0.05). Conclusion: The residualThin-prep cytology test specimen employed in HPV16E6/E7DNA feasibly. HPV16E6and E7gene could be as molecular marker in cervical cancer screening for these closelyrelated to cervical carcinoma in Xingjiang, and may has a broad prospect of clinical application for prediction disease progress and choose a more reasonable treatment ofcervical disease. |
