Angiogenesis is a crutial step in the growth and metastasis of cancers. This paper provided an overview on the mechanism of angiogenesis and presented the recent investigation of the anti-angiogenesis drugs. Above all,this paper introduced the anti-angiogenesis effects and the corresponding mechanism of some phenylpropanoids (include sinapine). Previous studies has revealed that Sinapine possessed anti-rumor activity and anti-angiogenesis activity,suggesting that anti-tumor angiogenesis may be one of the mechanisms of its anti-tumor effect,but its exact anti-tumor angiogenic activity and mechanism still need to be clarified.So,This study was designed to investigate the anti-tumor angiogenesis effect of sinapine and explored its mechanism of action in vitro,which,to some extent,might provede theoretical basis for the application of sinapine.The effect of sinapine on the supernatant of cultured S180cells induced angiogenesis was detected by Matrigel plug model.The explanted S180sarcoma model was built up to observe the effect of sinapine on the micro vessel density(MVD) and vascular endothelial growth factor(VEGF) expression of the tumor.To investigate the effect of sinapine on the proliferation,migration,invation of HUVEC,MTT assay and transwell were used.The effect of sinapine on the ability of tube formation of HUVEC was observed by culturing HUVEC on Matrigel.Gelatin zymography was performed to detecte the effect of sinapine on the MMP-2expression and activity of HUVEC. ELIS A assay was used to detecte the effect of sinapine on the VEGF expression of HUVEC and S180cells.The results indicated that sinapine can inhibit the angiogenesis of cultured S180tumor cells supernatant containing Matrigel plug and explanted S180sarcoma with a does-response relationship.The results also showed that sinapine significantly inhibited the angiogenesis related cell ability (include migration,invation,tube formation) and MMP-2,execept that, VEGF expression of HUVEC and S180cells was also down-regulated,which indicated that Sinapine can inhibit tumor angiogenesis,and inhibits the motility of endothelial cells through inhibiting MMP-2expression and activity of endothelial cell, down-regulating VEGF expression of endothelial cell and tumor cell might be the mehcanism of action of its anti-tumor angiogenesis effects.
