Research On The Function Of MiRNA-132in Transplantation Rejection In Vitro

Objective:To study the expressions of miRNA-132and VEGF-A gene during transplantation rejection in vitro, providing reference for controling transplantation rejection effectively.Methods:①Cells of EOMA were cultivated, and then mixed with T lymphocyte cells from the spleen of Balb/c mice to build model experiments of transplantation rejection in vitro. It was divided as EOMA group, EOMA+IL-2group and E0MA+IL-2+T lymphocyte cells group.②Cells were harvested at0,6th,12thand24th and the expression of miRNA-132was detected using the technology of TaqMan miRNA Assays.③The expression of VEGF-A mRNA was detected by qRT-PCR. The expression of VEGF-A protein was detected using immunocytochemistry and the EOMA’s proliferation was observed by MTT assay.Results:①Compared with EOMA group and EOMA+IL-2group, the expression of miRNA-132in EOMA+IL-2+T lymphocyte cells group showed a significant difference at the12th and24th hour (P<0.05). The expression of miRNA-132in EOMA+IL-2+T lymphocyte cells group was significant increase than other two groups (P<0.05). The expression of miRNA-132in the three groups had no difference at the0and6th hour (P>0.05).②Compared with EOMA group and EOMA+IL-2group,The expression of VEGF-A mRNA showed a significant difference in EOMA+IL-2+T lymphocyte cells group at the12th and24th hour (P<0.05); The expression of VEGF-A mRNA was significant increase in EOMA+IL-2+T lymphocyte cells group than other two groups at the12th and24th hour (P<0.05). The expression of VEGF-A mRNA in the three groups had no difference at the0and6th hour (P>0.05).③The expression of VEGF-A protein in EOMA+IL-2+T lymphocyte cells group was significant increase in EOMA+IL-2+T lymphocyte cells group than other two groups at the12th and24th hour (P<0.05). The expression of VEGF-A protein in the three groups had no difference at the0and6th hour (P>0.05).④Compared with EOMA group and EOMA+IL-2group, the EOMA cells’growth rate in EOMA+IL-2+T lymphocyte cells group was significant increase at the12th and24th hour (P<0.05), The growth rate in the three groups had no difference at the0and6th hour (P>0.05).Conclusions: During the early transplantation rejection, the expression of miRNA-132is upregulated, leading to the increase of VEGF-A, inducing the growth of EOMA, contributing to repairment of EOMA.
