Objective To Observe the expression changes of miRNA-221, miRNA-222in human glioma cell and tissue and discuss the correlation between the expression of miRNA-221/222and the radiosensitivity of human glioma.Methods Vitro U87, U251, A172, LN229glioma cell lines,Collect60cases of tissue samples of glioma from Tianjin huanhu hospital,all were confirmed by pathology..Extracted total RNA from U87, U251, A172, LN229glioma cell lines vitroed and60cases of glioma tissue collected through operation according to Trizol Kit; In the normal brain tissue for control, determine the expression of miR-221and miR-222through real-time PCR,.Check the radiosensitivity in each group of glioma cells by cloning experiments.60cases of glioma patients was treated with radiotherapy after surgery. Follow-up of glioma patients and analysis the radiotherapy in gliomas by MRI examination. In the ges for control,estimate glioma cell survival rate after X-ray irradiation (irradiation dose of2Gy) by cloning experiments. Verify the radiosensitivity of the glioma cell lines.Results1MiRNA-221and miRNA-222upregulation in glioma tissue.Compared with the control group and cell, miR-221, miR-222in glioma cell lines and tissue are highly expressed (P<0.05), Which is highly malignant gliomas of miRNA-221, of miRNA-222expression were significantly higher than low grade (P<0.05). WHO III and WHO IV glioma expression compared to the WHO Ⅱ or WHO Ⅰ was significantly higher (P<005). of miR-221and miR-222expression and the pathological grades of gliomas significantly correlated (P<0.05).2MiR-221, miR-222in glioma cell lines are highly expressed. Real time PCR confirmed: compared with the ges, miR-221, miR-222are highly expressed in the U87, U251, A172, Ln229glioma cells Department, and in the four glioma cellof miR-221/222expression in U87cells, in turn reduce the expression of the highest, of U251, A172, Ln229cells. Followed by X-ray irradiation after glioma cell viability (SF), and U87cell lines SF maximum, the radiation resistance is greater, of U251, A172, Ln229cell lines SF were decreased, radiation resistance is also in turn. MiRNA221/222 expression in Glioma cell was positively correlated with glioma SF after X-ray radiation.(r=0.673,P<0.01).3Low, highly malignant patients with postoperative radiotherapy and survival time comparison: Partial resection of the same level of glioma patients, MiR-221, low miR-222expression in patients with carcinoma after radiotherapy tissue mean reduced about92.5%, while miR-221, miR-222expression in patients after radiotherapy of tumor tissue average reduction degree only48.6%.In low-grade gliomas, in patients with other prognostic factors similar premise, miRNA-221and miRNA-222expression is low, the median survival of the patients was49.3±1.6months; and miRNA-221and miRNA-222expression in patients with high median survival period was23.8±0.7month. In patients with miRNA-221and miRNA-222expression and survival time of patients with correlation analysis revealed that r=-0.87, P<0.05. Similarly, in high-grade glioma patients, miRNA-221and miRNA-222expression is low, the median survival of the patients was22.3±0.9months; and miRNA-221and miRNA-222expression in patients with high median survival period was12.3±0.5month. In patients with miRNA-221and miRNA-222expression and survival time of patients with correlation analysis revealed that r=-0.63, P <0.05glioma patients with miRNA-221and miRNA-222expression and survival time of patients with negative correlation.Conclusions1MiR-221, miR-222in glioma cell lines and tissues are highly expressed, and with the increase of gliomas grade expression of volume increase. MiR-221and miR-222expression and malignant degree of gliomas were positively correlated (P<0.05).2MiR-221, miR-222in glioma cell lines are highly expressed. MiRNA-221/222expression and SF glioma cells is positively correlated.As the miR-221glioma cells, the expression of miR-222content increased, the radiation sensitivity is reduced.3Glioma tissue radiosensitivity reduced with miRNA-221/222expression decreased, MiRNA-221/222expression is negative correlation with the radiosensitivity survival time of patient...
