Objective:Some studies show that Crocin can inhibit the proliferation and induce the apoptosis of various of tumor cells.This study aims to investigate the effect of Crocin on malignant biological phenotype in human lung adenocarcinoma cell line of A549and its molecular mechanism.Method:Different doses of Crocin were added to the lung A549cell line to demonstrate the biological function.MTT was performed to reveal the inhibitory effect on cell proliferation,and flow cytometry was used to determine the apoptosis and cell cycle of the lung A549cell line after the treatment of Crocin.P53,bax and bcl-2were measured by RT-PCR.Results:There was a dose dependent inhibition of cell proliferation of human lung adenocarcinoma A549cell line by Crocin. The combination of Crocin and DDP/PMX can improve the chemosensitivity and induce the apoptosis more significantly of human lung adenocarcinoma cell line of A549(P<0.01). Cell cycle studies showed Crocin increasing the rate of G0/G1of A549cell cycle.Crocin could increase the expression of p53mRNA, down-regulate the expression of bcl-2mRNA,and had no influence on the expression of bax mRNA (P>0.05).Conclusion:Crocin can inhibit the proliferation and improve the chemosensitivity of lung A549cell line through inducing its apoptosis, changing cell cycle, increasing the expression of p53,and down-regulating the expression of bcl-2.
