Purpose:In order to further explore Chinese medicine on metabolic disorders of type2diabetes,an experiment on corn silk extract (CSE) was conducted to study its effect on blood sugar and fat metabolism in experimental type2diabetic rats.Methods:With high sugar and fat feeding, type2diabetic rats were made by small dose STZ injection which could damage β cell in pancreas islets. In addition to normal control group (normal group), the diabetic rats were randomized into5groups, diabetes model group (model group), metformin group, high dose CSE group,middle dose CSE group and low dose CSE group. After4weeks treatment, fasting blood glucose, triglyceride, total cholesterol,HDL-c and LDL-C were taken for test, meanwhile, pancreas, heart, liver and kidney were taken off for pathological test by immunohistochemistrystaining.Results:Comparisons among CSE groups, metformin group and model group after treatment:I. By treatment, fasting glucose in CSE groups and metformin group were lower than it in model group, with obvious dose-effect relation and statistical significance(P<0.05). Ⅱ.Triglyceride in CSE groups and metformin group were significantly different to it in normal group(P<0.05),meanwhile,which declined significantly by comparison with model group(P<0.01). By treatment, total cholesterol in high dose CSE group, middle dose CSE group and metformin group all fell obviously, with significant difference to model group(P<0.01). In high dose CSE group and metformin group, HDL-c rose and LDL-C fell by treatment, with significant difference to model group(P<0.05). Ⅲ. With light microscope and HE stain, it showed rich pancreas islets with normal configuration in normal group, decreased pancreas islet cells with degeneration and necrosis (++)in model group, clear structure of pancreas islets with mild degeneration and necrosis(+) in metformin group. In high dose CSE group and middle dose CSE group, morphology of pancreas were improved, especially, morphology of pancreas in high dose CSE group was close to which in metformin group. Protective action of high dose CSE on heart muscle was superior to metformin, and CSE may protect liver and kidney effectively and delay degeneration of liver cell and renal tubular epithelial cell.Conclusions:CSE is able to improve glucose and fat metabolism, reduce damage to pancreas, heart,liver and kidney in diabetic rats,and further delay the progress of diabetic complications.
