Objectives:Detecting the expression levels of E-cadherin, N-cadherin,β-catenin,vimentin and fibronectin in normal cervical tissue,cervicalintraepithelial neoplasia and cervical cancer, to observe the relationshipbetween them and cervical lesions, HPV infection. To explore the relationshipbetween EMT and cervical lesions,the association between HPV infection andthe occurrence of EMT, thereby providing a theoretical basis to find newtherapeutic targets for the control of cervical cancer malignant behavior.Methods:1Collected36cases of surgical resected cervical cancer,30cases ofcervical intraepithelial neoplasia and20cases of normal cervical tissueparaffin-bedded samples from Sencond Hospital of Hebei Medical Universitybetween2007and2011.All patients had no preopreative chemotherapy orradiotherapy, and underwent HPV testing.2SP immunohistochemical method to detect the expression ofE-cadherin,N-cadherin,β-catenin,vimentin and fibronectin in normal cervicaltissue,cervical intraepithelial neoplisia and cervical cancer.3Data was analyzed by SPSS19.0software, and the results were treatedby χ2test and Pearson Correlation analysis. P<0.05was statisticallysignificant.Results:1The expression of E-cadherin in cervical lesions.The positive expression rate of E-cadherin in20cases of normal cervicaltissue,10cases of CINⅠ,20cases of CINⅡ-Ⅲ and36cases of cervicalcancer was respectively80.0%,70.0%,45.0%,19.4%. The expression ratedifference in the four tissues had statistical significance (P<0.05). The positiveexpression rate of E-cadherin in cervical cancer with intravenous tumor thrombus group and without intravenous tumor thrombus group were5.6%,33.3%, and the difference between the two groups had statistical significance(P<0.05).E-cadherin in cervical cancer with lymph node metastasis group andwithout lymph node metastasis group positive expression rate was0.0%,33.3%,respectively,and the difference between the two groups hadstatistical significance (P<0.05).2The expression of N-cadherin in cervical lesions.The positive expression rate of N-cadherin in20cases of normal cervicaltissue,10cases of CINⅠ,20cases of CINⅡ-Ⅲ and36cases of cervicalcancer was respectively10.0%,20.0%,40.0%,75.0%. The expression ratedifference in the four tissues had statistical significance (P<0.05). The positiveexpression rate of N-cadherin in cervical cancer with intravenous tumorthrombus group and without intravenous tumor thrombus group were94.4%,55.6%, and the difference between the two groups had statistical significance(P<0.05).N-cadherin in cervical cancer with lymph node metastasis group andwithout lymph node metastasis group positive expression rate was93.3%,61.9%,respectively,and the difference between the two groups hadstatistical significance (P<0.05).3The expression of β-catenin in cervical lesions.The positive expression rate of β-catenin in20cases of normal cervicaltissue,10cases of CINⅠ,20cases of CINⅡ-Ⅲ and36cases of cervicalcancer was respectively80.0%,70.0%,40.0%,13.9%. The expression ratedifference in the four tissues had statistical significance (P<0.05). The positiveexpression rate of β-catenin in cervical cancer with intravenous tumorthrombus group and without intravenous tumor thrombus group were0.0%,27.8%, and the difference between the two groups had statistical significance(P<0.05). β-catenin in cervical cancer with lymph node metastasis group andwithout lymph node metastasis group positive expression rate was0.0%,23.8%,respectively,and the difference between the two groups hadstatistical significance (P<0.05).4The expression of vimentin in cervical lesions. The positive expression rate of vimentin in20cases of normal cervicaltissue,10cases of CINⅠ,20cases of CINⅡ-Ⅲ and36cases of cervicalcancer was respectively10.0%,20.0%,45.0%,72.2%. The expression ratedifference in the four tissues had statistical significance (P<0.05). The positiveexpression rate of vimentin in cervical cancer with intravenous tumorthrombus group and without intravenous tumor thrombus group were88.9%,55.6%, and the difference between the two groups had statistical significance(P<0.05). Vimentin in cervical cancer with lymph node metastasis group andwithout lymph node metastasis group positive expression rate was93.3%,57.1%,respectively,and the difference between the two groups hadstatistical significance (P<0.05).5The expression of fibronectin in cervical lesions.The positive expression rate of fibronectin in20cases of normal cervicaltissue,10cases of CINⅠ,20cases of CINⅡ-Ⅲ and36cases of cervicalcancer was respectively15.0%,30.0%,55.0%,80.6%. The expression ratedifference in the four tissues had statistical significance (P<0.05). The positiveexpression rate of fibronectin in cervical cancer with intravenous tumorthrombus group and without intravenous tumor thrombus group were94.4%,66.7%, and the difference between the two groups had statistical significance(P<0.05). Fibronectin in cervical cancer with lymph node metastasis groupand without lymph node metastasis group positive expression rate was100.0%,66.7%,respectively,and the difference between the two groups hadstatistical significance (P<0.05).6The correlation between HPV16,18infection and E-cadherin β-catenin,N-cadherin,vimentin,fibronectin expression.32cases had HPV16,18infection in20cases of normal cervicaltissue,30cases of CIN and36cases of cervical cancer, and the other54caseshad no such infection. In32cases HPV16,18infected tissues,9cases hadE-cadherin positive expression. Using the Pearson Correlation analysis, thecorrelation between HPV16,18infection and E-cadherin expression hadstatistical significance(P<0.05,r=-0.266).In32cases HPV16,18infected tissues,10cases had β-catenin positive expression. Using the PearsonCorrelation analysis, the correlation between HPV16,18infection andβ-catenin expression had statistical significance (P<0.05,r=-0.218).In32cases HPV16,18infected tissues,20cases had N-cadherin positive expression.Using the Pearson Correlation analysis, the correlation between HPV16,18infection and N-cadherin expression had statistical significance (P<0.05,r=0.322). In32cases HPV16,18infected tissues,19cases had vimentinpositive expression. Using the Pearson Correlation analysis, the correlationbetween HPV16,18infection and vimentin expression had statisticalsignificance (P<0.05,r=0.217). In32cases HPV16,18infected tissues,22cases had vimentin positive expression. Using the Pearson Correlationanalysis, the correlation between HPV16,18infection and vimentinexpression had statistical significance (P<0.05,r=0.236).Conclusion:1With the process of cervical lesions, the expression of E-cadherin、β-catenin gradually decreased, while the levels of N-cadherin、vimentin、fibronectin increased. According to this, conclusion was made that EMTdeveloped within the process of cervical lesions and facilitated the invasionand metastasis of the cervical cancer.2With the increasing infection rate of HPV16、18, the expression ofE-cadherin、β-catenin gradually decreased, while the levels of N-cadherin、vimentin、fibronectin also increased. A tentative inference on this result wasthat HPV might induce cervical epithelium developing EMT to result incervical lesions...
