Kashin-Beck disease(KBD) is an endemic chronic bone disease. The current studies show that:Low selenium in the environment, food fungal contamination, and organic acids in water are the three major possible causes.In this research, wheat samples were collected from the KBD area and strains of fungi were isolated from those samples to study the distribution of fungi in KBD region.This work provides a scientific reference for further study of its etiology and eventually control this disease.In this study,227strains of fungi were isolate from all samples and153strains were in diseases area and74in control area. Strains were identified by morphological feature and molecular biology methods to species and genus level, including:Fusarium.spp. Altrnaria.spp, Caphalosporium Corda. Humicola Traaen. Coccospora Wallr, Peniciuium spp, Oidiodendron robak. Aspergillus Mi-cheli ex Fr. Mucor Micheli, RhizopusEhrenberg. Saccharomycetacear(Meyen)Ree-ss, Glomerularia Peck. Ovulariopsis Pat.et Har, Dothichizalib. Acremoniella Sacc, Geotrichum LK, Diplococcium Grove. Monilochaetes Halst.ex Harter, Hel-mintho-sporium Lk.ex Fr, Nigrospora sphaerica(Sacc.) Mason. In totle21fungi genera were identified. The number of Fusarium.spp which isolate from disease rear was account for33.3%, but its number in control area was only account for13.5%. The number of Fusarium.spp in disease rear was significantly higher than it in control area. Those results indicate that there may some relationships between KBD and Fusarium.spp in decease rear.Additionally, some no-produced fungi stains were treated by promoting spore experiments.11stains were promoted to produce spores through different conditions in medium, light and temperature but7strains retain failed to produce spores. The results show that different medium have a large impact on production of spores than other conditions like light and temperature.Finally, the modified SDS method of DNA extraction was certained by compared other two methods. Six strains were identified by determined sequence of ITS region of DNA through molecular method and verify the result of morphological identification method.
