| Objective:To investigate the different expression of checkpoint kinase (Chk1) and multiple tumor suppressor1(p16) among triple negative breast cancer (TNBC), non-triple negative breast cancer (non-TNBC) and adjacent normal tissues of breast cancer (BC); investigate the expression of Chk1, p16expression in TNBC, moreover to investigate the relationship of clinical pathology in TNBC; illuminate the relationship and clinical manifestation of Chk1, p16expression in TNBC.Methods:Immunohistochemical ElivisionTM plus method was used to detect the expression of Chk1, p16expression in47TNBC tissues,39non-TNBC tissues and26adjacent normal tissues of BC. Analysis the different expression of Chk1, p16in47TNBC tissues; to illuminate the clinical manifestation of Chk1, p16expression in TNBC.Results:1. The positive rate of Chk1in47TNBC tissues is68.09%(32/47); in39non-TNBC tissues is46.15%(18/39); and with the positive rate of19.23%(5/26) in adjacent normal tissues of BC. The positive rate of Chk1in47TNBC tissues is higher than non-TNBC (P<0.05); the positive rate of39non-TNBC tissues is higher than the adjacent normal tissues of BC (P<0.05); and the positive rate of Chk1in47TNBC tissues is higher than the adjacent normal tissues of BC too (P<0.05).The positive of Chk1with rates of â…¡ degree54.55%and â…¢ degree92.31%in47TNBC tissues respectively. The expression and its level of Chk1protein is correlated with histological grade (P<0.05). But There was no significant difference for patients age,size of tumor and lymph node metastases,menstrual status,pathologic types and clinical TNM (P>0.05).2. The positive rate of p16in47TNBC tissues is27.66%(13/47); in39non-TNBC tissues is48.72%(19/39); and with the positive rate of84.62%(22/26) in djacent normal tissues of BC. The positive rate of p16in47TNBC tissues is lower than non-TNBC (P<0.05); the positive rate of39non-TNBC tissues is lower than the adjacent normal tissues of BC (P<0.05); and the positive rate of p16in47TNBC tissues is lower than the adjacent normal tissues of BC too (P<0.05).The positive of p16with rates of â…¡ degree45.45%and â…¢ degree7.69%in47 TNBC tissues respectively.The expression and its level of p16protein is correlated with histological grade (P<0.05). But there was no significant difference for patients age, size of tumor and lymph node metastases,menstrual status, pathologic types and clinical TNM (P>0.05).3. The expression of Chkl, p16in47TNBC tissues have inverse correlation in their protein level (P<0.05).Conclusions:1. Chkl is overexpression in TNBC and non-TNBC,but especially in TNBC; pi6is underexpression in TNBC and non-TNBC, but especially in TNBC.2. The expression and its level of Chk1protein is correlated with histological grade, but there was no significant difference for patients age, size of tumor and lymph node metastases, menstrual status, pathologic types and clinical TNM; the expression and its level of p16protein is correlated with histological grade, but there was no significant difference for patients age, size of tumor and lymph node metastases, menstrual status, pathologic types and clinical TNM.3. The expression of Chkl, p16in47TNBC tissues have inverse correlation in their protein level.4. The underexpression of p16and the overexpression of Chkl may play a significant role in the incidence and development of BC, especially in TNBC. The expression level of Chkl, p16protein correlated with histological grade, so combination of these factors will be more helpful for treating BC or TNBC. |
