The Effects Of Direct Intra-articular Injection Of Adenovirus-fasl On Apoptosis Of Synoviocytes In Adjuvanpt Arthritis Rats

Rheumatoid arthritis (RA) is a systemic autoimmune disorder characterized bysynovial tissue hyperplasia with infiltration of various inflammatory cells leading tocartilage and bone destruction. RA disability high, seriously affecting the quality oflife.Currently,RA is not curable.Gene therapy may provide a safe and effectivetreatment for the patients.In this study,we direct delivered the adenovirus containingFasL gene into adjuvanpt athritis rats through intra-articular injection and observedthe apoptosis of snoniocytes.This experiment establishes a solid base for further studyof gene therapy for rheumatoid arthritis.Objective：Observe the effects of direct intra-articular injection of Ad-Fasl on apoptosis ofsnoniocytes in Adjuvanpt Athritis Rat.Method:1.Construction of recombinant adenovirus carrying FasL gene.2.Models of adjuvanpt athritis in SD rats were induced by.intracutaneous injection of Freund’s complete adjuvant in The right hind paw.3.Adenovirus carrying FasL gene were delivered into adjuvanpt athritis rats byintra-articular injection.4.Clinical markers of adjuvanpt arthritis rats such as body weigh﹑arthritisindex were observed.5.Pathomorphologycal changes of Synovial tissue of adjuvanpt arthritis ratswere observed.6.Apoptotis of synoviocytes of adjuvanpt arthritis rats were observed byTUNEL. Result:1.Body weigh of adjuvanpt arthritis rats transfected with FasL gene was higherthan transfected with Lacz.2.Reduction of arthritis index of adjuvanpt arthritis rats transfected with FasLgene was higher than transfected with Lacz.3.Significant reduction in Synovial cell proliferation and infiltration ofinflammatory cells was found in adjuvanpt arthritis rats transfected with FasLgene.4.Apoptotis index of adjuvanpt arthritis rats transfected with FasL gene washigher than normal rats transfected with FasL gene﹑normal rats transfectedwith Lacz or adjuvanpt arthritis rats transfected with Lacz.Conclusion：Through intra-articular injection of Ad-FasL can enhanced apoptotis index ofsynoviocytes as well as improved AI of rats.
