| Objective: To research the effects of the myeloid differentiation factor-88(MyD88)-mediated toll-like receptor9(TLR9) signal transduction pathways on tumor necrosisfactor (TNF-α) and Interleukin-6(IL-6) and the reference between ulcerative colitisactivity and the TNF-α/IL-6by detecting the TLR9, MyD88, TNF-α and IL-6inperipheral blood of humans with ulcerative colitis (UC) and normal humans.Methods: The expression levels of TLR9mRNA and MyD88mRNA in peripheralblood were detected by RT-PCR; The IL-6and TNF-α protein in serum in differentgroups were detected by enzyme-linked immunosorbent assay (ELISA). The expressionlevels of TLR9, MyD88, IL-6and TNF-α among the patients with active UC, remissionpatients and control group were analyzed to compared with the defferent grouos.Results:1.RT-PCR results: The expression levels of TLR9mRNA and MyD88mRNAin peripheral blood lymphocytes of patients with active UC was higher than that inremission and normal control group, and the difference was statistically significance(p<0.05). There were no significant difference (p>0.05) between the remissionpatients and the control group.The mRNA expression of TLR9and MyD88in severeUC group were higher than that in the mild or moderate group (p<0.05). The correlationof mRNA of TLR9and MyD88were analyzed by Pearson. There were a positivecorrelation (r=0.806, p<0.01) between TLR9mRNA and MyD88mRNA.The correlation of mRNA of TLR9and MyD88and the UC activity levels wereanalyzed by Spearman analysis. There were a positive correlation in them (TLR9rs=0.810p<0.01, MyD88rs=0.799p<0.01.).2. ELISA results: The TNF-α and IL-6concentration in serum of active UCgroup and the remission goups was statistically difference comparing with the controlgroup (p<0.05). Their concentration between in the remission goups and the controlgroup was no statistical significance (p>0.05). Their concentration in the severe UC group was higher than that in the mild or moderate group (p<0.05). The correlationcoefficient of the concentration of TNF-α and IL-6was was0.834(p<0.01). The correlationcoefficient of aomng the concentration of TNF-α and IL-6and UC activity level were0.820(TNF-α p<0.01) and0.879(IL-6p<0.01).Conclusion:1. The relationships among TLR9, Myd88, IL-6and TNF-α in the peripheralblood in UC patients are significantly positive correlation, and TLR9may contribute toMyd88mRNA expression by resulting the release of inflammatory cytokines.2. TLR9, MyD88, IL-6and TNF-α expression level in UC patients aresignificantly higher than that in remission and normal control group, and the severity ofUC is positively correlated to the factors, suggesting that it related to the activity of theUC disease. |
