Objective To evaluate the effect of lentiviral vector-mediated FOXO1overexpression on expression of aquaporin9in the human hepatoma cellline HepG2.Methods Lentiviral overexpresssion vector of human FOXO1pWPI-FOXO1was constructed. Lentiviral vector was used to infect HepG2cells.GFP positive cells were sorted by FACS for further culture. Real TimePCR and Western blot analysis were carried out to confirm overexpressionof FOXO1in HepG2cells. After that, expression change of AQP9inHepG2cells was also detected by Real Time PCR and Western blot analysis.Results Lentiviral expresssion vector of human FOXO1wassuccessfully constructed. FOXO1mRNA and protein level in FOXO1overexpression cells was significantly higher than that in cells infected byLentiviral vector only expressing GFP and no infected cells(P<0.05).AQP9mRNA and protein level in FOXO1overexpression cells also significantlyincreased(P<0.05). Conclusions We concluded that lentiviral vector-mediated FOXO1overexpression exactly effect expression of aquaporin9in the humanhepatoma cell line HepG2. The expression of AQP9in HepG2cells isincreased by lentiviral vector overexpressing FOXO1,which suggestFOXO1may play a role in regulating AQP9expression to some extent,butthe specific mechanism is necessary to be further discussed.
