Objective:To investigate stargazin protein expression(coded by gene CACNG2,voltage-dependent calcium channel y-2subunit) in the patients of cortical dysplasia(CD) with intractable epilepsy (IE) and in order to explore the probable relationship between stargazin expression and CD/IE.Methods:Firstly we had collected the cortex from6persons,3cases from CD with IE and3cases from normal person, then used Gene Chip technology and analyzed the results, and found the differential expressed genes. Then We expanded the sample capacity to20cases and selected gene(stargazin) possibly related to epilepsy. Using polymerase chain reaction(PCR), immunohistochemistry, and western blot(WB) analysis, stargazin expression was examined in ten human neocortex samples from patients who underwent surgery to treat the CD with IE(The case group) and ten histological normal neocortex from patients who underwent cerebral trauma and brain gliomas(the control group).Results:(DWe had discovered485different genes with Gene chip, Among which the Diffscroe of350genes were less than-20, The diffscroe of135genes were more than20;â‘¡RT-PCR analysis revealed the expression of stargazin mRNA(CACNG2) was increased in the cortex of humans with CD and IE contrast to control group at the same period (P<0.05);â‘¢Immunohistochemical results showed that expression of stargazin appeared in the cytoplasm and cell membrane of cortical neurons; the expression of stargazin was obviously increased in the cortex of humans with CD and IE contrast to control group(P<0.05);â‘£Western-Blot results demonstrated the expression of stargazin protein level was obviously increased in cortex of humans with CD and IE(P<0.05) contrast to control group at the same period(P<0.05).Conclusion:â‘ There are485different genes from the neocortex of patients with CD and IE by Gene chip, Among which135genes were upregulated,350genes were downregulated.â‘¡The upregulation of stargazin in cortex of patients with CD and IE may play a role in participating the process of epilepsy or/and forming intractable epilepsy.
