Objective: TPX2(Targeting for Xklp2) is a recently discovered genewhich involves in cell mitosis and is mutually related to proliferation,apoptosis, invasion and metastasis of various cancers. shRNA plasmidtargeting TPX2gene was constructed in the first part of the research andthe effect of proliferation and invasion on human lung adenoma A549cellswas observed in the second part. It provides datas and thought for clinicaltreatment of lung adenoma.Methods: Specific shRNA plasmids for TPX2gene were constructedand were stably transfected into A549cells by lipofectamine2000.Expression level of TPX2gene, the proliferation-related genes(CDK4and CyclinD1) and the invasion-related gene (MMP2) wereobserved by RT-PCR and western blot.MTT measured its effects onproliferation. The cellular invasion was tested by Transwell chambers.Results: The TPX2shRNA plasmid for human TPX2gene wasconstructed successfully. It inhibited the expression of the mRNA and theprotein of TPX2gene remarkably and the inhibition rates were73.8%and 80.2%(P<0.05). The mRNA expression of the proliferation-related gene(CDK4and CyclinD1) decreased and the inhibition rates were76.69%and67.28%(P<0.05). The expression of the invasion-related gene (MMP2)was reduced by58%(P<0.05).Proliferation and invasion of A549cellswere decreased markedly. It was statistically significant.Conclusion: The growth of A549cells was inhibited. Proliferationand invasion were decreased apparently. The results indicated TPX2genemight be a potential target for lung adenoma therapeutics in the future.
