Objective: To observe the effect of5-aza-2’-deoxycytidine on cell growth ofHepG2and the expression of Beclin1, and to investigate the potential mechanism ofits anti tumorigenesis.Methods: Human hepatocellular carcinoma HepG2cells were treatedwith(0,0.4,1.6,6.4,25.6,102.4)μmol/L5-aza-CdR for24h,48h and72h, with0μmol/L5-aza-CdR as negative control, Cell growth inhibition was assayed by MTTmethod.Morphological changes of HepG2were observed by phase contrastmicroscopy under different drug concentrations in different time periods.To detect theeffect of5-aza-CdR on expression of mRNA and protein in tumor suppressor geneBeclin1by RT-PCR and Western blot.Results: It was demonstrated by MTT assay that the rates of inhibition were(1.1±0.36)%,(10.80±2.27)%,(35.21±3.25)%,(49.20±3.28)%,(75.70±2.41)%,(84.30±3.31)%respectively after treatment with various concentration0μmol/L,0.4μmol/L,1.6μmol/L,6.4μmol/L,25.6μmol/L,102.4μmol/L5-aza-CdR for72h.5-aza-2’-deoxycytidine could inhibit HepG2cell growth in a concentration and timedependent manner. It was demonstrated by RT-PCR and Western blot that5-aza-CdRincreased Beclin1expression of mRNA and protein. The results showed that after72hours in102.4μmol/L5-aza-CdR intervention, Beclin1expression of mRNA andprotein increased significantly. Difference was statistically significant compared withthe control group(P<0.01).Conclusions:1.5-aza-2’-deoxycytidine can inhibit the proliferation of humanHepatoma cells HepG2.2.5-aza-2’-deoxycytidine can increase the Beclin1expression of mRNA and protein in human Hepatoma cells HepG2in a concentration and time dependent manner.
