Objective: To explore the Synergistic actions of Celecoxib combined withPitavastatin on proliferation and apoptosis on colon cancer SW480cell line, and it’smolecule mechanism and pathway.Method: Colon cancer cell SW480was cultured In vitro, cell of exponentialphase of growth is used to experiment. With different concentrations of Celecoxib,Pitavastatin and Celecoxib conbined with Pitavastatin of a period of time, MTT assaywas used to analysis the inhibition rates and proliferation of SW480; Flow cytometrywas used to detect cell apoptosis; the expression of mRNA of PAkt, COX-2wasdetermined by RT-PCR; the expression of pritein of PAkt, COX-2was determined byWestern-blot assay.Results: Both of Celecoxib and Pitavastatin inhibited colon cancer cell SW480proliferation, and into a time and dose dependent; the inhibition of combinedtreatment group(Celecoxib and Pitavastatin) was significantly higher than inhibitionby Celecoxib or Pitavastatin individually (P<0.05), Kim Jong methods shows q>1.15,suggested a synergistic effection of two drugs on inhibition of colon cancer cellproliferation, and increased with prolonged duration of action; Flow cytometryshowed that, Celecoxib (45μM), Pitavastatin(25μM) and the combined group treatedon SW480cells at48h, the apoptosis rates of the three groups were10.28%±2.84%,14.32±4.59%,31.54±4.38%, apoptosis of the three groups were significantly higherthan the control group (P<0.05), apoptosis of combination group was significantlyhigher than the single drug group(P<0.01). Celecoxib and Pitavastatin, and theircombination signifycantly inhibited the expression of mRNA and protein of PAkt andCOX-2in SW480cells(P<0.05), combined treatment group had significantdifferences when compared with the single drug group(P<0.05).Conclusions: Celecoxib and Pitavastatin inhibit the proliferation of colon cancerSW480cells in a concentration-dependent and time-dependent positive, and promoteapoptosis in colon cancer SW480cells, One of the possible mechanisms of apoptosis is to inhibit the expression of PAkt, then to inhibit the parhway of PI3K/Akt, and inthe same time inhibit the expression of COX-2.There was some synergistic effectionsof the combined use of Celecoxib and Pitavastatin on the inhibiting of proliferationand inducing apoptosis in colon cancer SW480cells.
