Objective:Useing Phage Display Technology in vivo to pan the short peptidespecifically combined with ovarian cancer cell A2780, and it may becomes to thecarrier of targeted drags of cancer in order to guide a new way of targeted therapy ofthe ovarian cancer.Method: To establish the nude mouse tumor model. The phages was injectedinto the nud mouse through the tail vein and paned for three rounds, got the enrichedphage which binded to the short peptide. The phage titer of tumor, heart, liver andbrain tissue assayed and then tested in the method of Immunohistochemistry andELISA. The phage DNA was extracted and then sequenced, analysed the sequence bybioinformatics. The peptides was synthesised and detected the inhibition of ovariancancer cell A2780by MTT.Result: Nude mouse tumor model was sueeessfully established. After threerounds of panning, phage recovery was titered, which binding to the tumor tissue is3.4×103times of the Liver tissue,1.4×106times of the heart tissue,1.3×106times ofthe brain tissue. Immunohistochemis-try showed that the peptide binding to the tumortissue enriched significantly but the heart, liver and brain tissue didn’t. ELISA showedthat of all the10clones P/N>2.1has8. It illustrated this8clones have a high affinityfor A2780. We obtained the peptides pecifically binding to the phage and sequenceingit is PHLATLF. And validated the inhibition of ovarian cancer cell A2780by MTT.Conclusion: The peptide PHLATLF specifically combined with ovarian cancercell A2780was successfully screened and indentified by ELISA andImmunohistochemis-try. In order to make the futhur research of the peptide coupledwith chemotherapeutics next. And esteblish the foundation of new targeted druges ofovarian cancer.
