Pancreatic cancer is one of the most lethal abdominal malignancies of which the earlydiagnosis and therapy are extremely difficult, with an overall five year survival rate ofonly1%to5%. Currently, the widely used biomarkers for pancreatic cancer,such asCA19-9, CEA, CA50, etc, show unsatisfactory sensitivity and specificity.Therefore,better biomarkers for early diagnosis are desperately needed. This projectattempts to screen differentially expressed proteins in serum samples fromwell-diagnosed patients of pancreatic cancer, chronic pancreatitis,benign pancreaticdisease and normal control by2D-DIGE and MALDI-TOF-MS technology and toidentify and quantify the differentially expressed proteins by LC-MS/MS technology.Differentially expressed proteins of pancreatic carcinoma were be identified.Objective(1) To detection the repeatability and stability of2D-DIGE together withMALDF-TOF-MS screening serum biological markers of pancreatic cancer;(2) Analysising serum of pancreatic cancer patients and control groups by usingcomparative proteomic methods to looking for he potential specific candidatemarkers of pancreatic cancer, and providing the theory basis and the new way ofthinking for the pathogenesis and early diagnosis of pancreatic cancer and earlydiagnosis.Methods(1) Proteins differentially expressed in serum samples from40patients of pancreaticcancer,10chronic pancreatitis,10benign pancreatic disease and40normal controlwas screened by2D-DIGE; (2) Significant differentially expressed protein spots were identified byMALDI-TOF-MS to find differentially e xpressed proteins associatedwith early diagnosis of pancreatic cancer.ResultsThree differentially expressed proteins of pancreatic cancer: complementcomponent3(C3)、Transthyretin(TTR) and Apolipoprotein E(APOE) were Successfulidentified.Conclusion(1) The method of screening serum biological markers of pancreatic cancer by2D-DIGE together with MALDF-TOF-MS has a good repeatability and stability;(2) The finding of inflammation mediated factor C3revealed that nflammation may beclosely related with the development and development of pancreatic cancer.(3) The identified protein TTR and ApoE in this study may be as specific serumbiomarkers of pancreatic carcinoma.
