Express Of MicroRNA-23b And MicroRNA-200b In Plasma Of Systemic Lupus Erythematosus Patients And It’s Clinical Significance

Objective:To research the expression level of microRNA-23b(miR-23b), MicroRNA-200b(miR-200b) in the plasma of patients with systemic lupus erythematosus (SLE).and to analyze the relationship between expression level and clinical manifestations, such as disease activity level, renal involvement., then, preliminary study the clinical significance of MicroRNA-23b(miR-23b), MicroRNA-200b(miR-200b) in SLE.Method:The plasma of50SLE patients,22healthy people was collected. The expression level of miR-23b, miR-200b was detected by Real-time fluorescence quota polymerase chain reaction (QRT-PCR),all SLE patients were Classificated into two groups according to SLEDAI scoring methods:SLE active phase group(28cases) and non-SLE active phase group(22cases),with renal involvement situation,it were classificated into lupus nephritis group and non-lupus nephritis group.and detect the clinical indexes such as BR、UR、ESR、CRP、Immune full set of inspection24hours total urine protein (24h-UTP)、features of lung image、renal biopsy and histopathology examination meanwhile. One-Way ANOVA and Spearman’s correlation analysis were used to evaluate the relationship between expression level and clinical manifestations.Result:①miR-23b and miR-200b were turned out to be relative to SLE by conjointed analysising of3database analyse(miRBase、PicTar、 TargetScan).②The expression level of miR-23b, miR-200b in plasma is significant lower in active stage than in remission stage(P<0.01) and in healthy control (P<0.01);③The expression level of miR-23b and miR-200b in plasma is lower in Lupus nephritis patients than no kidney involvement group(P<0.01) and healthy control(P<0.01).④The expression level of miR-23b is negative correlated to ESR, complement in blood serum, SLEDAI score, and chronic kidney disease index (CI)., miR-200bConclusions:①The expression level of miR-23b, miR-200b in plasma may be one of the indicators of the disease activity of SLE.②The miR-23b may be associated with the occurrence and prognosis of nephritis Lupus nephritis.
