ObjectiveThe effects of intravous transplantation of human cord blood mononuclear cells (HCMNCs) combined with perindopril on inflammatory responses and Interleukin-6(IL-6) in the myocardium in rabbits with acute myocardial infarction were investigated.MethodsThe model of acute myocardial infarction(AMI) was established by ligation of the left anterior coronary artery (LAD). The60healthy rabbits of the model built successfully were divided into four groups randomly,each group consisted of15animals.(1) Control group:gavaged with saline2ml daily for4weeks, and saline0.5ml via ear vein injection in24h after operation.(2)Perindopril group:gavaged with perindopril2mg/kg.d daily for4weeks, and saline0.5ml via ear vein injection in24h after operation.(3)Transplantation group:gavaged with saline2ml daily for4weeks, and3×107of HCMNCs labeled with GFP in normal saline0.5ml via ear vein injection.(4) Combined Group:gavaged with perindopril2mg/kg.d daily for4weeks, and3×10’ of HCMNCs labeled with GFP in normal saline0.5ml via ear vein injection in24h after operation. At1,2,4weeks after the treatment, cardiac function indexs such as left ventricular fractional shorting(LVFS) and ejection fraction(LVEF) were performed by echncardiography. The white blood cell count in the myocardium stained with HE were determined by light microscopy. The expression of IL-6in the cardiac tissue was detected by immunohistochemistry. The number of transplanted cells in the myocardium was found by GFP positive cells counted with fluorescence microscopy.Results(1).The heart function indexs such as LVEF and LVFS were significant improved in the Perindopril group, transplantation group and combined group compared to control group after transplantation at1week,2week,4week respectively(P<0.05). The cardiac function was improved significantly in transplantation group was better than that in Perindopril Group (P<0.05), and most significantly in the combined group (P<0.05) among the three treatment groups.(2).The white blood cell count and expression of Interleukin-6(IL-6) protein were decreased significantly in the Perindopril group, transplantation group and combined group compared with the control group (P<0.05). Compared with Perindopril group, The white blood cell count and expression of IL-6protein was lower in transplantation group (P<0.05), and lowest in the combined group (P<0.05) among the three treatment groups.(3).GFP-positive cells were found to be distributed in the peri-myocardial infarction area in the both transplantation and combined groups at1,2,4weeks post-transplantation, and while the number of the GFP positive cells in the combined group was found more than that in the transplantation group (P<0.05).Conclusions1. Intravenous transplantation of human cord blood mononuclear cells can survive in the myocardium of rabbits with AMI and improve heart function. The possible mechanism may be involved to inhibit inflammatory responses and expression of IL-6after myocardial infarction induced by HCMNCs.2.The combination of perindopril with intravous transplantation of HCMNCs in the treatment of rabbits with AMI can increase survival rate of transplanted cells, further enhance improvement of the heart function. The possible mechanism may be related to the combined treatment, which can inhibit the myocardial inflammation and the expression of interleukin-6more effectively. |
