Objective:To investigate the effects of VB2, a noval lignanoid from the acetoacetate extract of Vitex negundo seed on the proliferation and apoptosis and the express influence on mTOR/4EBP1mRNA signal pathway in human choriocarcinoma JEG-3cell lines in vitro.Methods:The inhibitory effects of different concentrations of VBE-2and different time JEG-3cells was assessed by methyl thiazoly1tetrazo1ium(MTT)assay; flow cytormetry was used to analyze apoptosis after used the different concentrations of VBE-2; the expression of mTOR and4EBP1mRNA was determined by RT-PCR.Results:1:VB2inhibited the proliferation of JEG-3cells.The inhibitory rate of JEG-3cells growth, which were cultured with different concentrations of VB2(2.5,5,10,20,40,80,160umol/L) after indicated time (24、48、72hours) was from (6.34±0.41)%to (85.89±0.81)%. and it was appeared the significantly positive correlation with the dose (P<0.05);2:VB2induced the apoptosis of JEG-3cells.0、5、10、20umo1/1VB2increased the rare of JEG-3cell apoptosis in vitro from (7.26±1.42)%to (35.85±1.24)%,after48hours. it was appeared clear dose dependent (P<0.05);3.0、5、10、20umo1/1VBE-2could down regulate the mRNA levels of mTOR and4-EBP1after48hours, which was a significant negative correlation (P<0.05).Conclusion:VB2can restrain choriocarcinoma cell JEG-3proliferation, induction of its apoptosis. This effect may relevant to VB2inhibit JEG-3cell mTOR/4E-BP1mRNA expression.
