PART ONE The effects of estrogen on Estrogen Receptor α36in Human Osteoblasts-like CellsObjective To examine the role of estrogen in stimulating ER-a36in MG-63osteoblasts-like Cells.Methods Cultivating MG-63in vitro,when the cells was impacted by different kinds of media concentration of estrogen, the mRNA level of ER-a36was examined by using Quantitative real-time PCR(QRT-PCR).The protein level of ER-a36was evaluated by Western Blot.Results E2at10-10mol/L or greater concentrations reduces ER-a36expression in MG-63;Conclusions Estrogen can effect the expression of ER-a36in MG-63. PART TWO The effects of mechanical stimulation on Estrogen Receptora36in Human Osteoblasts-like CellsObjective To examined the role of mechanical stimulation in stimulating ER-a36in MG-63osteoblasts-like Cells.Methods MG-63osteoblasts-like Cells were subjected to mechanical tension/compress strain with four-point bending system (ZL01256849.X) at0.2Hz frequency and2000μstrain for0,1,3,6,and12hours respectively.The mRNA level of ER-a36was examined by using Quantitative real-time PCR(QRT-PCR).The protein level of ER-a36was evaluated by Western Blot.Results Mechanical stimulation time-dependently decreased both ER-a36mRNA and protein levels after continuiously stretch for1h,and reached a minimal response afer continuiously stretch for3h.Conclusions Mechanical stimulation decreased the expression of ER-a36in MG-63osteoblasts-like Cells.
