Study On The Pathogenicity Of Excessive DM8909in Female Mice Reproductive Tract

Objective:To analyse the pathological characteristic of mice infected by the only Ureaplasma urealyticum serotype1(Uu1) or the only Lactobacillus DM8909and the two mixed microorganism,In the purpose of mastering more knowledges about the pathogenicity of the two kinds of micro-organism.Method:156oestradiol-treated adult female mice of strain ICR(Institute of Cancer Researcch) were randomly divided into6groups.There were36mice in each group A-B,24mice in each group C-E,while mice of control group(group F) inoculated sterile medium. Uul with the concentration of107Copy/g was given to the group of A while Uul with the concentration of10’Copy/g and DM8909with the concentration of1012Cfu/ml were given to the group of B.DM8909with the106,109,1012were inoculated intravaginally to the groups of C,D,E. They were detected DM8909from vaginal secretions by quantitatively live bacterial counting at1,3,7,14,21,28days of postinoculation randomly.Then they were necropsied at1,3,7,14,21,28days of pstinoculation randomly,to detect Uu from cervical secretions by fluorescent quantitative polymerase chain reaction(FQ-PCR) and(or) detect DM8909from cervical secretions by quantitatively live bacterial counting,observing tissues of cervix,endometrial,fallopian tube by light microscope and cevial mucosa,endometrium by scanning election microscope.Results:(1)The total positive rates of Uul in two experimental groups showed a significant difference(p<0.05),A were52.8%(19/36),B were27.2%(10/36) And the positive rate declined as the days of postinoculation lasting.The highest positive rates of two experimental groups were100.0%(6/6),83.3%(5/6) at1d after inoculation,but the secretions didn’t persisit at28d.All mice of control group were negtive.(2)There was statistical difference in the quantity of Uu among experimental groups(p<0.05) in day1,3-Their peak level were1.99×107,9.33×104reached at1d, the FQ-PCR values reduced as the days of postinoculation lasting.(3)Vaginal quantitative value of DM8909:the average vaginal value of control group is5.13×103Cfu/ml, the content of inoculated intravaginally lactobacillus in group(B-E) of mice was in a dose-dependent increase.There appeared vaginal value climax at3d or7d after the inoculation.There was significant difference in the vaginal quantity of DM8909in B and E experimental groups in any time.(4) Cervical quantitative value of DM8909:The peak level was8.13×107Cfu/ml reached at3d in group B,While their peak level were1.86×104,4.68×106,1.32×109Cfu/ml reached at7d in group C-E, There was significant difference in the cervical quantity of DM8909in B and E experimental groups in any time.The quantitative value in the vagina was more1or2logs than the value in the cervical.Maybe the effection of the lactobacillus in the vagina was higher than in the cervical in the mice.(5)The total pathogenicity rates of two groups were significant different at3-28d,A were53.3%(16/30), B were13.3%(4/30),(P-0.001<0.05),none of control group was positive.Conclusions:(1) Lactobacillus DM8909might reduce the infection rate of Uu on the cervical epithelial cells.DM8909had inhibitory effect on Uu growth.(2)The Ureaplasma urealyticum serotype1(Uu1) inoculated intravaginally probably had depressant effect on DM8909.(3)Lactobacillus DM8909can stick the cervical epithelial cells.(4)Lactobacillus DM8909had better sticky effect on the vaginal epithelial cells than on the cervical epithelial cells.(5) The excessive lactobacillus DM8909had no apparent destruction to the vaginal or cervical of mice the same as the Ureaplasma urealyticum serotype1(Uu1) to the mice.
