| Aims:this paper tries to study if Korean medical recipe of "qianjinwenwutang" has the influence on glycated hemoglobin content which is animal model with Diabetic Nephropathy (DN)Method:this experiment was taken in Physiology research office of medicine department of Yanbian University and animal experimental center from May,2010-December,2010.The experiment choose Wistar male rats130only, the weight is about180g-250g, Among them,122rats cut off the left kidney;8rats which is normal control group do not have the operation. After7days’wound healing, the rat with nephrectomy will inject Streptozocin,STZ(55mg/kg) which is dissolved by Citric acid buffer. However, normal control group will inject equivalent Citric acid buffer. After molding72hours, using Fundus venous blood sampling method to test FGB and Scr; multiply urine output per minute by formula Ucr/Scr multiply per minute’s urine output to calculate Ccr, taking24h urine to test PUV and Ucr. The big rat with GLU>16.7mmol/1,24h PUV≥80ml, Ccr≥1.5ml/min is successful in building Diabetic nephropathy model. After centrifugal cryopreservation, the rest of the blood in the urine samples to be measured.40male wistars which are successful in building model were divided to5groups by random number method and randomization:UUO group (8mice), positive control group (8mice), high-dose of " qianjinwenwutang " group (8mice), middle-dose of " qianjinwenwutang " group (8mice), low-dose of "qianjinwenwutang" group (8mice) and add normal control group. The total is6groups. Experiment will test weight, HbAlc,24h proteinuria and Blood Urea Nitrogen (BUN) after finished molding. lOml/kg.d distilled water was poured to UUO group and normal group by gastric perfusion,1mg/kg.d nuohelong was poured to positive control group by gastric perfusion, and10g/kg.d high-dose of "qianjinwenwutang",5g/kg.d middle-dose of " qianjinwenwutang ",2.5g/kg.d low-dose of "qianjinwenwutang" group were poured separately by gastric perfusion. Experiment will test each group’s weight, the weight of kidney and renal index,24h PUV,24h proteinuria, fasting blood glucose,glycated hemoglobin, Scr,Ucr and BUN. Usingformula Ucr/Scr to multiply every minute’s urine, and then calculate Ccr. Every datawas showed by means士standard deviations(x土s), used sotfware of GraphpadPrism5.0to deal with the Analysis of variance test. Lastly,尸<0.05was considered tohave statistical significance.Experimental results: body weight and right renal index:â‘ The model group ofinjecting drug, positive control group,"qianjinwenwutang"large dose group,"qianj inwenwutang" middlâ€e dose group,“qianj inwenwutang low dose groupwere signiifcantly lower than those of control group (P <0.01); the weight of“posiitve control group,"qianjinwenwut"anglarge dose group, qianjinwenwutang"middle dose group,"qianjinwenwutang" low dose group were significantlydecreased. It was no significant difference comparing with the model group (P>0.05). After injecting drug, the right kidney index of the model group, positive controlgroup,"qianjinwenwutang" large, middle, low dose group,were significantly higherthan that of normal control group (P <0.01); compared with the model group,positive control group,"qianjinwenwutang "large, middle, low dose group has nosignificant difference (P>0.05).â‘¡24h urine volume: the24h urine volume of modelgroup was significantly less than control group,and the difference was obvious (P<0.01); after injection, comparing with the model group,the positive control group,"qianj inwenwutang " large, middle, low dose group urine volume was obviouslydecreased,the difference was significant P <0.01).â‘¢24h urine protein quantitative:atfer making model,24h urine protein content of model group was higher than that ofcontrol group, the difference was significant (P <0.01).24h urine protein content ofpositive control group,“â€qianj inwenwutang large, middle, low dose group wassignificantly lower than the model group, there was obvious difference (P <0.01).â‘£Blood glucose: atfer modeling, the model group glucose was higher than thecontrol group, there were significant difference (P <0.01). Atfer injecting drug,blood gluc“ose of the positive control group,"qianjinwenwutang" large, middle, lowdose group were significantly lower than that in the model group, and it had significant difference (P <0.01).⑤The glycosylated hemoglobin: Atfer modeling,glycosylated hemoglobin of the modle group of was significantly higher than that incontrol group, it had significant difference (P <0.01, P <0.05). Atfer injecting drug,comparing with the model group, the glycosylated hemoglobin of positive controlgroup,"qianjinwenwutang" large,middle, low dose group had significant difference(P <0<.01, P0.05),â‘¥The BUN, Scr,Ccr: Atfer modeling,the BUN of model groupwas higher than that of the control group, there were significant difference (P <0.01);after injecting darg, the BUN of positi“ve control group,"qianjinwenwutang" large,middle, low dose group were lower than that in the model group, it had significantdifference (P <0.05,P <0.01).Atfer buiding the model,Scr, Ccr of model groupwere significantly higher than those in control group with significant difference (P<0.01); atfer injecting,Scr, Ccr of positive control group,"qianjinwenwutang" large,middle, low dose group were all lower than that in the model group with signiifcantdifference (P <0.01).Conclusions: l.In this experiment the program of intraperitneal injecting STZ atferunilateral renal resection has successfally establish the DN medeling.2.Korean medical recipe of “qianjinwenwutang†can reduce DN rat’sGLU and HbAlc, also can control the level of GLU.3Korean medical recipe of "qianjinwenwutang " can improve thegeneral condition of DN rats.4.Korean medical “recipe of qianjinwenwutangcan reduce DN rat’sScr, content of Urea nitrogen, decrease Urine protein deduction, and improve DN ’ratsrenal function. |
