Modern pharmacological studies have shown that purifier which has been used for the treatment of hepatobiliary diseases has high immune activity and has the functions of ictercus-retreating, gallbladder-availing, liver-cell-protecting and digestion-aiding. Furthermore, bear gallbladder powder has the advantages of small toxicity and little dose. At present, coarse powder, capsules (including compound capsules) and tincture of bear gallbladder are made through direct draining of bear bile by people at home and abroad. Draining bear gallbladder powder has been studied in Medical School of Yanbian University since1991. The injected bear bile powder is a kind of purifier bear bile powder which is refined through fat-freeing, decolourizing, membrane-separating, freeze-drying. Pharmacological research shows that this injected bear bile powder has the function of blood circulating, relieving convulsion and spasm and clearing head,so this powder has been used to treat people suffering cerebral thrombosis, myocardial infarction or other thrombotic diseases. The research of methodology has been carried out to determine the content of the active ingredients such as both combined and free of ursodeoxycholic acid and chenodeoxycholic acid.The content of combined ursodeoxycholic acid and combined taurocholate chenodeoxycholic acid in injected bear bile powder was determined by HPLC. Chromatorgraphy was performed on a YMC-Pack ODS-A (5-C18-AR-Ⅱ) column,and the mobile phase was methanol:NaH2PO4(72:28,PH4.4), and the flow rate was0.8mL/min,and column temperature was30℃,and the wavelength of detection was210nm. The determination method of combined taurocholate ursodeoxycholic acid and combined taurocholate chenodeoxycholic acid in injected bear bile powder has been studied.The content of free ursodeoxycholic acid and free chenodeoxycholic acid in injected bear bile powder was determined by HPLC. Chromatorgraphy was performed on a Cosmosil ODS (5c18-AR-Ⅱ) column, and the mobile phase was cetonitrile:NaH2PO4(45:55, PH3.0), and the flow rate was1.0mL/min, and column temperature was20℃, and the wavelength of detection was205nm. The determination method of free ursodeoxycholic acid and free chenodeoxycholic acid in injected bear bile powder has been studied.Experimental results show that: The content of unhydrolyzed combined taurocholate ursodeoxycholic acid and combined taurocholate chenodeoxycholic acid were42.07%and11.73%respectively, the overall content was53.80%.The content of free ursodeoxycholic acid and free chenodeoxycholic acid which were alkaline hydrolysis and extracted with ethyl acetate,were39.96%and11.15%,and the overall content was51.11%, equivalent to combination,the content after hydrolysis were higher than that of unhydrolyzed.The resules indicated that the method of determination of hydrolyzed was better than that of unhydrolyzed and it is worthy of promotion and application.
