Objective:The experimental observation of puerarin combined Methylcobalamin treatment of peripheral neuropathy in diabetic rats by measuring the sciatic nerve tissue of diabetic rats to oxidative stress indicators (SOD, MDA) and Na+-K+-ATPase activity and so to explore the possible role of mechanism.Method:In the early exploration STZ streptozotocin modeling based on the best modeling dose (55mg/kg) by intraperitoneal injection system may diabetic rat peripheral neuropathy model62, and then use of puerarin and Acobalamin treatment medication, alone and in combination group administration, were randomly divided into model group,alone mecobalamin group, single-puerarin group, two-drug combination group (n=15). Dynamic determination of rat body weight and blood glucose observed after the administration of at least8weeks DPN blood glucose, blood glycosylated hemoglobin content; observe the sciatic nerve pathological changes, determination of sciatic nerve conduction velocity, sciatic nerve tissue superoxide dismutase SOD and malondialdehyde MDA of Na+-K+-ATPase in order to explore the Pueraria factors to improve the polyol pathway, inhibit the role of non-enzymatic glycosylation and improve microcirculation and nerve function, methylcobalamin nutrient nerve tissue, and anti-apoptosis effect and to explore its mechanism.Result:Using STZ injection model, can make blood glucose levels of diabetic rats increased from about4.8±0.21mmol/1to23±0.93mmol/1, P<0.01, there was a significant difference, combined with sciatic nerve pathological and usually rats a little over performance that caused by diabetic peripheral lesions (DPN) model.The puerarin prime contrast administration model group can lower blood sugar, increase SOD activity and decreased MDA content, P<0.05, significant difference.Methylcobalamin contrast administration model group can improve the DPN in rat sciatic nerve conduction velocity (P<0.05), the role of nutrition of the sciatic nerve, improve nerve morphological changes.4two combined group compared single-drug group, to retain the single use of biochemical indicators to improve the trend, observed combination of apparent harm.Conclusion:Successfully prepared the diabetic rat peripheral neuropathy models (DPN).2This article does not Methylcobalamin significant effects on blood glucose levels, puerarin and the combined group a significant effect on blood sugar drops, reveals the main mechanism of the two drugsthe improvement of blood sugar levels. Further confirmed Methylcobalamin nerve tissue protective effect, reducing the pathological damage of diabetic DPN in rat sciatic nerve, the mechanism may be the protection of neuronal cell bodies, to prevent irreversible degeneration, is the key to recovery after peripheral nerve injury. Therefore, given exogenous to promote nerve regeneration after peripheral nerve injury substances may reduce the retrograde degeneration of the cell body. Puerarin can improve the DPN in rat sciatic nerve tissue SOD activity and decreased MDA content, and effective inhibition of oxidative stress changes in diabetic rats. The Pueraria peripheral neuropathy in diabetic rats activity is superior to the combination of factors and Methyl cobalamin alone, has a neuroprotective effect, the mechanism may improve nerve polyol metabolism and reduce nerve inflammation and other factors.
