Objectives: To investigate drug resistance and drug resistance genes andgenotypes of MRSA isolates from nasopharyngeal carriage of healthy adults andsputum of hospital respiratory infection patients in Nanjing area of China.Methods: A total of19MRSA strains isolated from nasopharyngeal samples ofhealthy adults and51MRSA strains isolated from sputum of hospital respiratoryinfection patients were collected. Antibiotic susceptibility testing was detected byK-B methods. Multiplex PCR was used to detect the methicillin resistance gene mecA;the aminoglycoside resistance gene aacA-aphD; themacrolide-lincosamide-stretogramin B resistance gene ermA and ermC; and thetetracycline resistance gene tetM and tetK. The genotypes of SCCmec weredetermined by multiplex PCR. Molecule groupling for the MRSA strains weredetermined by pulse field gel electrophoresis(PFGE).Results:All MRSA strains were sensitive to vancomycin, teicoplanin and linezolid.The resistant rates of all strains to many kinds of drugs were above90%. MecA genewas detected in all MRSA strains. The positive rates of aacA-aphD, ermA, ermC,tetM, and tetK genes in healthy adults and hospital infection patients were94.8%,100.0%,21.0%,100.0%,10.5%and98.0%,96.1%,19.6%,96.1%,23.5%,respectively. Among the19and51isolates of MRSA,SCCmec III accounted for 78.9%and88.2%,SCCmec II accounted for21.1%å'Œ11.8%respectively. SCCmecI,IV and V strains were not discovered in our study. PFGE typing results showed9types for MRSA, some MRSA strains in healthy have close genetic relationship withthat isolated from sputum of hospital respiratory infection patients.Conclusion: Multiple resistance of MRSA are both serious in healthy adults andhospital infection patients, and the positive rates of drug resistance genes are veryhigh, which should be paid high attention to. The dominant epidemic strain of MRSAin healthy and hospital infection patients were SCCmec type III. Nosocomial MRSAstrains may transmitted into community and colonized in healthy persons.
