Objective:To study the effects of inflammatory cytokines and brain edema in rats ofacute cerebral ischemia and reperfusion with different electroacupuncture stimulationintensity. Neurological deficit score (NSS) and the infarct area were observed in eachgroup. The levels of inflammatory cytokines, TNF-α, IL-1β, IL-10and the content oftissue water in brain in the peripheral blood serum were analysed. Explore the therapeuticeffect of acupuncture strength of the best EA on rats of cerebral ischemia and reperfusion.Methods:8week-old SD rats were randomly divided into3groups: the normal group,10;the sham operation group,10; model group,30. Established the middle cerebral arteryocclusion (MCAO) and reperfusion model in the model group, then divided its into threegroups: the model control group, the weak EA group, the strong electro-acupuncturegroup. Did the acupuncture6hours after the ischemia-reperfusion (I/R). Did the NSS,collected blood of abdominal aortic and the brains after24hours. The brain of2rats inevery intervention group were frozened section, and the infarct area was observed withTTC staining. Meanwhile, the brain water content were detect with wet and dry method.detected the levels of TNF-alpha, IL-1β, IL-10in the Serum, and the statistical analysiswas done for every index. Results:1. varying degrees nerve function deficit and the righteye Horner’s certificate were dicovered in the model group, and the normal group andsham group does not appear above signs, and varying degrees of cerebral infarction weredicovered after the TTC staining;2. Compared with the normal group, the levels ofinflammatory cytokines, TNF-α, IL-1β, IL-10and the content of tissue water in brain wereno statistically significant differences (P﹥0.05) in the sham operation group, and itsincrease significantly (P﹤0.01, P﹤0.05) in the model control group;3. Compared withthe model group, the levels of TNF-alpha, IL-1β decreased significantly (P﹤0.05, P﹤ 0.01), the levels of IL-10and the content of tissue water in brain were no statisticallysignificant differences (P﹥0.05) in the weak EA group. But, in the strong EA group, thelevels of TNF-alpha, IL-1β and the content of tissue water in brain decreased significantly(P﹤0.01, P﹤0.05), while, the levels of IL-10increased significantly (P﹤0.01);4.Compared with the weak EA group, the levels of TNF-alpha, IL-1β decreasedsignificantly (P﹤0.05), the levels of IL-10and the content of tissue water in brain wereno statistically significant differences (P﹥0.05). Conclusion: EA can protect the rats inthe acute cerebral ischemia reperfusion by reducing TNF-alpha, IL-1β and brain tissuewater content and increasing IL-10levels of the serum. the efficacy are differenceamong the different intensity EA, strong stimulation is superior to the weak stimulation.
