A Intial Exploratory Study Of The Coagulation Dysfunction In Rats In Agkitrodon Acutus Venom-induced

Objective: through the way of intraperitoneal injection to build the ratsdisease model of agkitrodon acutus venom infection, and then accord-ing to themonitoring results of hematological indexes and microscopic observation inlung tissue of rats, to explore the change of coagulation function andmicrocirculation after the agkitrodon acutus venom infection,in order toprovide theoretical basis for the clinical treatment.Methods: receie30health clean level Sprague-Dawley rats which areweighed180-200g, randomly (random number table) divided into infectedvenom24hours group and another contrastive group. The venom soluti-on with the mixed agkitrodon acutus venom powder and saline in proportion of3:1, infected group rats intraperitoneally inject1.75mg/kg venomsolution, and the contrastive group rats intraperitoneally inject the samedoes saline.(1). Observe the rats of2groups poisoning performance successively aft-er injected venom up to the experiment finished.(2). Each rats of2groups abdominal aortic venous blood samples3.0mlafter injected venom, and the blood will be encapsulated into EDTA ant- icoagulant-tube and PT anticoagulant-tube immediately, gently shake theblood to avoid forming blood clots.(3). The rats were then killed were anatomized to take the right upper l-obe tissues samples after taken the blood sample, and then put the tissu-es into formalin stored under the normal temperature and avoids the lig-ht to keep.(4). Blood sample and tissue samples are mounted specimen in2hours,and then send to the First Affiliated Hospital of Guangxi Medical unive-rsity clinical laboratory and pathological laboratory.Results:(1). The infected group rats intraperitoneally injected venomsolution have change of behavior in carrying degrees after30minutes, b-ut the change of behavior are returned to normal in12-24hours. No si-gnificant abnormality in behavior with the contrastive group. The contras-tive groups has no obvious behavior change during the experiment period.(2). The results of hematology detection showed that the PLT and FIB inblood decrease obviously in the infected group rats, but RBC, HB, PT a-nd clotting protein(Ⅷ、Ⅸ、ⅩⅠ) have no significant abnormality with c-ontrastive group.(3). No lung microthrombosis is observed through histo-pathological examination.Conclusion: Initial speculation, agkitrodon acutus venom can cause th-rombocytopenia in body, to keep body in a state of without-fibrin(FIB), t-hus appeared the symptom of clinical bleeding as a hallmark,but not ca-use DIC. Overall, it is a clinical situation of consumptive coagulation dia-orders in body.
