Endoscopy has experienced four stages which include Rigid endoscopse, semi-flexed endoscopes, semi-flexed endoscopes, electronic endoscopes since the emergence. It’s widely used in the diagnosis and treatment in many areas of ear, nose, throat, bladder, etc. Endoscopic technology and other subjects also had more extensive contacts with the development of medical and health undertakings. For example, the ultrasound technology was used in the endoscope system. Some scholars use fluorescence and endoscopic technology combine to create a fluorescence cystoscopy. As a new method of a laser field of medicine, PDT is now active in the treatment of various tumor diseases by the unremitting study from domestic and foreign scholars. PDD technology has become one of the major methods for early tumor diagnosis recent years, because of its advantages of high sensitivity and high detection rate. Fluorescence diagnosis is widely used in the diagnosis of cancer of the oral cavity, hypopharynx and larynx, gastrointestinal tract, urinary system which have achieved good results.Objects:Although scholars have used endoscopy technology in PDD or PDT of tumor. But there is not such single system with both PDD and PDT function was reported. The purpose of this paper is to improve the structure of the traditional rigid endoscope to build up a endoscopy system with both PDD and PDT function, and test system performance and effectiveness.Methods:1. Design the pre-experiments on cellular level. Find a simple method by using Image Processing Toolbox from matlab software, which is used to determine the perfect time for PDD in the later animal experiments2. Improve the structure of the traditional rigid endoscope. Design the endoscope which contain PDD and PDT function. Purchase of the light source, imaging, control, laser system which are required. Complete the installation and commissioning of the system. Enable it to achieve real-time monitoring and save images in the diagnosis and treatment3. Implante tumor cells to BALB/c mice, buld up the tumor animal model. Complete the experiment with the endocope system after tumor formed.Results:1. The endoscope system has had video recording, display and storage capabilities which are required for the experiment. The structural design of the endoscope system can export the different wavelengths of laser which required by PDD and PDT. And ensures that the output power. It also achieves the connection of CCD system and PC. video recording and image capture can be control by PC, which for post-production processing and analysis.2. Studies have shown that the method based on image processing technology to research the process of photosensitizer absorbed by cells is feasible. The image processing of laryngeal cancer cell fluorescence picture by Sobel operator and Otsu operator is very successful. The experimental results reflect the extent of absorption of the photosensitizer by the cells. The histogram which obtained by experimentcan be used to inferr a reasonable time frame for post-experiment3. This experiment tests the PDD function of the endoscope system. The common digosic function and PDD function have been test in the dark environment which simulate in vivo condition. PC can acquisition, display and save the vedio signal from CCD system.Conclusion:The animal experiment proves the diagnosis and treatment of the alaser power transmission, video recording, displaying, saving of the image and the fluorescence excitation and other functions of the endoscope system to be qualified. It ill reach a higher accuracy of diagnosis and treatment effect after further investment and improvement, and eventually be applied into clinical.
