The main clinical features of syndrome of blood stasis are pain,mass andecchymosis.Modern research shows that syndrome of blood stasis hasbusiness with the abnormal of blood circulation,tissue and cell metabolism;obstacle of immunologic function;blood hi-viscosity state;platelet activation,adhesion and aggregation;thrombosis;many kinds of pathological andphysiological changes.Show that syndrome of blood stasis has closelyinterrelated with disturbance of blood circulation.For more than one thousand years,syndrome of blood stasis has beencured by Chinese herbal medicine under the guidance of traditional Chinesemedicine science. Chinese herbal medicine has been affirmed with a clear rolefor promoting blood circulation and removing blood stasis and has lessnoxious side effects for a long-term practical application.For some decades now,researchers have screened out many componentsfrom Chinese herbal medicine which have strong effect of promoting bloodcirculation and removing blood stasis.There are at least seven kinds ofmaterials:flavonoids,ferulic acid,sulfide,terpenoid,alkaloids,glycoside,steroidset cetera.Recently,some researchers report that Panax notoginseng saponions(PNS)can improve fibrinolytic activity,reduce fibrinogen,inhibit plateletaggregation,improve the abnormal of hemorheology,prevent thrombosis,improve microcirculatory disturbance and delay atherosclerosis formation.The root of Thladiantha Dubia (RTD)is the ripe root of cucurbitaceousplant Thladiantha Dubia Bunge. Long-term application of RTD decoctionamong the folk has proved that RTD has effects of promoting bloodcirculation and clearing away the heat-evil and expelling superficial evils, withless toxicity and side-effects, suggesting that RTD is a kind of Chinese herbalmedicine deserving further research. Experimental studies had shown that alcohol extract of ThladianthaDubia root(AETR) has effects of entending the bleeding time and clottingtime in mice with acute blood stasis.In this experiment, AETR can remarkablyimprove the abnormal of hemorheology,entend the clotting time,inhibitplatelet aggregation in rats with acute blood stasis; improve the auriclemicrocirculatory disturbance in mice with acute blood stasis.Part1Study on promoting blood circulation and removing blood stasisaction of alcohol extract of Thladiantha Dubia Root in rats with acuteblood stasisObjective:To study the influences of AETR on hemorheology, clotting time andplatelet aggregation in rats with acute blood stasis,to show the evidences ofAETR can promote blood circulation and remove blood stasis.Methods:1.The model of rats with acute blood stasis was produced by injectingAdrenaline hydrochloride and bathing in ice water. Detect hematocrit, Kvalue,plasma viscosity,whole blood viscosity and evaluate the effect of AETRon hemorheology in rats with acute blood stasis.2.The model of rats with acute blood stasis was produced by injectingAdrenaline hydrochloride and bathing in ice water.Detect PT,TT and APTTand evaluate the effect of AETR on clotting time in rats with acute bloodstasis.3.The model of rats with acute blood stasis was produced by injectingAdrenaline hydrochloride and bathing in ice water. Detect platelet aggregationrate and evaluate the effect of AETR on platelet aggregation rate in rats withacute blood stasis.Results:1.The results showed that HCT of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR(42.1±2.4,41.2±2.0,39.9±2.2(x±s,%)) were lower than themodel group(45.2±1.7(x±s,%)),there was statistical significance(P<0.01). The K value of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR (3.51±0.40,3.35±0.32,3.16±0.33(x±s)) were lower than the model group (4.08±0.33(x±s)),there was statistical significance(P<0.01).The plasma viscosity of ratsin0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR (1.20±0.07,1.19±0.06,1.18±0.03(x±s,mPa·s)) were lower than the model group(1.35±0.03(x±s,mPa·s)),there was statistical significance(P<0.05or P<0.01).The bloodviscosity(low shear) of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR(10.259±1.242,10.552±1.218,9.361±1.187(x±s,mPa·s)) were lower than themodel group(12.032±1.376(x±s,mPa·s)),there was statistical significance(P<0.05or P<0.01). The blood viscosity(middle shear) of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR (5.220±0.611,5.191±0.578,4.915±0.730(x±s,mPa·s)) were lower than the model group(5.887±0.406(x±s,mPa·s)),there was statistical significance(P<0.05or P<0.01). The blood viscosity(highshear) of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR (4.185±0.706,4.046±1.017,3.906±0.604(x±s,mPa·s)) were lower than the model group(4.748±0.662(x±s,mPa·s)),there was statistical significance (P<0.05or P<0.01).2. The results showed that PT of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR (19.60±2.51,20.45±2.92,20.69±2.44(x±s,s)) were longer thanthe model group(15.45±4.04(x±s,s)),there was statistical significance (P<0.05or P<0.01). The TT of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR(20.91±3.39,21.06±2.44,21.86±3.03(x±s,s)) were longer than the modelgroup(16.86±1.48(x±s,s)),there was statistical significance(P<0.05orP<0.01). The APTT of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR(17.38±2.01,18.80±1.60,19.66±2.44(x±s,s)) were longer than the modelgroup(16.39±1.22(x±s,s)),there was statistical significance(P<0.05orP<0.01).3. The results showed that platelet aggregation rate of rats in0.7g·kg-1,1.4g·kg-1groups of AETR(43.72±6.99,40.01±9.38(x±s,%)) were lower thanthe model group(50.22±11.59(x±s,%)),there was statistical significance(P<0.05). Conclusion:AETR can improve the abnormal of hemorheology,entend the clottingtime,inhibit platelet aggregation in rats with acute blood stasis,show thatAETR has the effect of promoting blood circulation and removing bloodstasis.Part2Influence of alcohol extract of Thladiantha Dubia Root on auriclemicrociculation in mice with acute blood stasisObjective:To study the influences of AETR on auricle microciculation in mice withacute blood stasis, to show the evidence of AETR can promote bloodcirculation and remove blood stasis.Methods:The model of mice with acute blood stasis was produced by injectingAdrenaline hydrochloride and bathing in ice water. Observe the dismeter ofarteriole and venula,the number of micrangium crossing net patefaction andevaluate the effect of AETR on auricle microciculation in the mice with acuteblood stasis.Results:The results showed that in20minutes the dismeter of arteriole of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR (44.38±4.17,45.63±3.20,46.25±3.53(x±s,μm) were higher than the model group(38.13±2.58(x±s,μm)),there was statistical significance(P<0.05or P<0.01).In20minutes thedismeter of venula of rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups ofAETR(78.13±3.72,81.88±2.58,83.75±2.32(x±s,μm) were higher than themodel group(66.88±2.58(x±s,μm)),there was statistical significance(P<0.05or P<0.01). In20minutes the number of micrangium crossing net patefactionof rats in0.35g·kg-1,0.7g·kg-1,1.4g·kg-1groups of AETR(4.25±0.46,4.50±0.53,4.63±0.74(x±s,n) were higher than the model group(3.63±0.52(x±s,n)),therewas statistical significance(P<0.05or P<0.01).Conclusion: AETR can obviously improve the auricle microcirculatory disturbance inmice with acute blood stasis.
