Objective Adipose-derived stem cells (ADSCs) hold great promise for repair andregenerating burn wounds by producing growth factors, but proinflammatorycytokines such as IL-8released by ADSCs would potentially deepen the wound andinhibit healing. The aims of our research were to identify the effects ofN-Acetyl-L-cysteine (NAC) on the proliferation, death and Interleukin-8(IL-8)production of ADSCs.Methods ADSCs were isolated from adipose tissue, then they were cultured in normalculture condition. Surface molecular expressions of ADSCs were analyzed by flowcytometry and immunofluorescence technique. In presence or in absence of NAC,ADSCs proliferation was examined using CCK-8kit and cell death was evaluated byflow cytometry analysis. After that, we detected IL-8mRNA expression by rt-PCRand protein production by ELISA.Results ADSCs exhibited a fibroblast-like morphology. In flow cytometry, ADSCsexpressed CD90, CD44, CD13and CD105, and were lacking in CD14, CD71, CD34and CD45. In immunofluorescence, ADSCs expressed CD44and CD105, and werelacking in CD34and CD45. We observed that cell proliferation in NAC-treated groupwas significant1.53folds to control group, the OD of treated group were1.217±0.165on day8, which were significant1.53folds compared to the non-NAC group of0.794±0.031(P<0.05). The apoptosis rate of ADSCs decreased by55.4%comparedwith control groups (P<0.01), from3.22%to1.65%, and the necrosis rate decreasedby48.8%(P<0.01), from7.65%to3.41%. Additionally, the IL-8mRNA expressiondecreased to46.2±8.7%of control group, and the IL-8protein production inNAC-treated group was269.34±43.58pg/ml, which was9.98±0.57%compared to the 2690.26±371.09pg/ml in control group (P<0.01).Conclusions ADSCs can be isolated from adipose tissue by collagenaseⅠdigesting.We believe that NAC might be helpful in burn wound repair and regeneration, bystimulating the proliferation of ADSCs, inhibiting cell death and suppressing IL-8production.
