Objective: This experiment was used to investigate the effect and possiblemechanism of knockdown of Sema4D gene in lung cancer A549cells.Methods: Sema4D short interfering RNA oligonucleotides were transfected into lungcancer A549cells. Real time PCR was performed to test the interferential effect ofSema4D gene on mRNA level. Immunofluorescence was used to detect the locationand expression of Sema4D protein in A549cells. Protein level of Sema4D andAKT/P-AKT were tested by western blot. MTT was performed to test the inhibitionrate of A549cells of knockdown Sema4D gene. The migration and invasion of A549cells were detected by transwell.Results: The expression of Sema4D gene in lung cancer A549cells was decreased onmRNA and protein level, and located in the cytolymph of A549cells. The inhibitionrate of Sema4D gene was69%on mRNA level by Sema4D siRNAoligonucleotides-C. The proliferation, migration and invasiveness of lung cancerA549cells were decreased by Sema4D siRNA oligonucleotides-C in24hours afterinterfere.Conclusions: The expression of Sema4D gene affects the proliferation, migration andinvasiveness of lung cancer A549cells, maybe have a significant relation with PI3K/AKT pathway.
