| Objective To investigate the clinical and pathological features of primary central nervous system diffuse large B cell lymphoma (PCNSL-DLBCL) and to measure the microvessel density within the tumor tissue for the purpose to study the biological and clinical significance of microvascular proliferation in PCNSL-DLBCL.Methods1. Twenty-three cases of PCNSL-DLBCL were collected from the archives of our department from2008to2011and its clinical and pathological features were analyzed. Ten cases of non-CNS-DLBCL and eleven cases of glioma (WHO Grade â…¡-â…¢) were selected as the control groups.2.Using the immunolabelling of CD31and CD34to demonstrate the endothelia of microvessels and depicting the distribution of microvessels, the microvessel density(MVD) in tumor tissue were examined. The differences between groups were evaluated statistically.3. All data were analyzed by SPSS13.0. The independent samples t-test was used. P<0.05was considered as with significant difference.Results1. Within our PCNSL-DLBCL cases,22of23cases being with the normal immune status, one case was with defective immune function. Patients’ages were from22to73years old with median number of50years. The ratio of male to female was14:9in our series.2. Using the immunolabelling of CD31, the MVD(vessel numbers/mm2) values were obtained from PCNSL-DLBCL, non-CNS-DLBCL and glioma groups respectively, as following:35.71±14.68ã€33.33±15.62ã€54.60±26.49. Using the immunolabelling of CD34, the MVD(vessel numbers/mm2) values were also obtained from PCNSL-DLBCL, non-CNS-DLBCL and glioma groups respectively, as following:41.15±19.68ã€33.97±16.49ã€69.43±39.45. No significant difference of MVD was found between PCNSL-DLBCL and non-CNS-DLBCL groups (P>0.05). However, significant difference of MVD was found between PCNSL-DLBCL and glioma groups (P<0.05). The PCNSL-DLBCL was with less microvessels distribution compared with that in gliomas (WHO Grade â…¡-â…¢). Moreover, there was no significant difference between the GCB subtype and non-GCB subtype groups (CD31:40.19±15.15ã€33.52±14.73, P>0.05; CD34:48.17±23.25ã€37.41±17.19, P>0.05).ConclusionPrimary central nervous system lymphoma occurs mainly in elderly. Diffuse large B-cell lymphoma is as the main histopathological type with immuno-subtypes of GCB (34.78%) and non-GCB (65.22%) in our series. The microvascular angiogenesis in PCNSL-DLBCL may not explain its different biological behavior compared with non-CNS-DLBCL. However, lower microvascular angiogenesis in PCNSL-DLBCL, compared with glioma, implicating its critical role leading different clinical pathological features. |
