Expression And Clinical Significance Of X-linked Inhibitor Of Apoptosis Protein (XIAP) And Protease Nexin-1(PN-1) In The Bone Marrow Tissue Of Leukemia And Lymphoma

Objective1. To investigate the expression and clinical significance of the X-linked inhibitor ofapoptosis protein (XIAP) and protease nexin-1(PN-1) in bone marrow biopsy tissue ofleukemia and lymphoma.2. To investigate the role of protease nexin-1(PN-1) on the X-linked inhibitor ofapoptosis protein(XIAP) in the protate cancer cell-PC-3ML cell line with increasedRecombinant Human Serpin E2/PN1, we hope that we can find out the influence of tumor.Methods1. The expression of XIAP and PN-1was detected by immunohistochemical assay in thebone marrow biopsy tissue from10cases of acute myeloid leukemia,13lymphoma and9patients without hematological malignancy.2. By the addition of Recombinant Human Serpin E2/PN1to protate cancer cell-PC-3ML cells, the expressions of XIAP and PN-1were detected by Real-timeQuantitative PCR and western blot.Results1. The level of XIAP in the bone marrow tissue from acute myeloid leukemia andlymphoma was1.88±1.79,1.50±1.84,respectively.They are higher than those from controlcases without hematological malignancy.And p=0.0024,P=0.001, respectively.2. The level of PN-1in the bone marrow tissue from acute myeloid leukemia andlymphoma was1.56±0.82,1.26±1.02, respectively. They are higher than those from control cases without hematological malignancy.And P=0.0024；P=0.001, respectively.3. Significantly decreasing expression of XIAP gene in protate cancer cell-PC-3ML cells,which were added of Recombinant Human Serpin E2/PN1, can be detected by Real-timeQuantitative PCR and Western blot.Conclusion1. The increased expression of XIAP and PN-1protein may contribute to the progressionof acute myeloid leukemia and lymphoma, which may be relevant to its role as regulating thebiological reaction of differentiation and development and apoptosis.2. protease nexin-1(PN-1) down-regulates X-linked inhibitor of apoptosis protein (XIAP)expression in protate cancer cell.
