ObjectiveTo investigate the role of microRNA-21in icaritin-induced apoptosis of hepatic stellate LX-2cell and possible mechanism involved.Methods1.Detection of the apoptotic cells by4’,6-diamidino-2-phenylindole and flow cytometry.2.Real-time polymerase chain reaction was used to detect microRNA-21expression changes.3.Transient transfection with pre-microRNA-21and anti-microRNA-21to regulate microRNA-21expression.4. Western blot was used to examine the protein expression of Bcl-2and Bax.Results1.Icaritin dose-dependently increased the apoptotic of LX-2cells (P<0.05)2.Icaritin dose-dependently decreased the expression of microRNA-21in LX-2cells (P<0.05)3.Overexpression of microRNA-21reduced icaritin-induced LX-2cells apoptosis (P<0.05),but inhibition of microRNA-21expression increased icaritin-induced LX-2cells apoptosis(P<0.05).4.Icaritin downregulated the protein expression of Bcl-2and upregulated the protein expression of Bax in LX-2cells.MicroRNA-21partly reversed the protein expression of Bcl-2and Bax induced by icaritin.ConclusionsMicroRNA-21mediates icaritin-induced LX-2cells apoptosis.
