Liao Asarum commonly used in traditional Chinese medicine, the active ingredient is a volatile oil, volatile oil containing mainly3,5-dimethoxy toluene, safrole and methyleugenol. In the Liao Asarum active ingredient research mainly uses gas chromatography-mass spectrometry analysis of the fingerprint technology used active ingredient in the Liao Asarum, one-component quantitative analysis using gas chromatography or liquid chromatography. However, due to the different origin of the Liao Asarum, the large differences in the quality and composition of complex and vulnerable to counterfeiting and adulteration, and the complexity of the Chinese medicines, often its pharmacological multiple components together, so the content of a single component as the evaluation of drug Liao Asarum indicators, there are some limitations, can not effectively control the quality of traditional Chinese medicine (TCM). In this paper selected the Liao Asarum main component3,5-dimethoxy toluene, safrole and methyleugenol content as a basis for evaluation of the quality of the Liao Asarum herbs.In this paper, the gas-liquid continuous extraction method to extract Liaoning volatile oil, further separation of impurities through the self-made silica gel column purification to get the3,5-Dimethoxytoluene, safrole and methyl eugenol mixture as a reference. And the establishment of the application of mixed reference standards, Liao Asarum acetone in the volatile oil by GC or HPLC method for simultaneous determination of the Liao Asarum3,5-dimethoxy-toluene, safrole and methyl eugenol content. Methodology to verify the GC method for the determination of the Liao Asarum3,5-dimethoxy toluene, safrole and methyl eugenol of content, precision RSD%, respectively0.95,1.25,1.85, reproducibility RSD%were1.90.1.60,1.64, sample recovery%, respectively99.16,94.99,102.57; HPLC method for the determination of the Liao Asarum3,5-dimethoxy-toluene, safrole and methyl eugenol, precision RSD%, respectively,1.77,1.07,1.39, reproducibility RSD%, respectively1.98,2.52,2.21%of the recoveries95.07,94.30,98.26. In this paper, a lot of science, detailed experimental data, fully demonstrating the application of mixed reference standard determination of the Liao Asarum3,5-dimethoxy toluene, safrole and methyleugenol content of scientific and practical to determine the3,5-dimethoxy toluene, safrole and methyl eugenol content in the Liao Asarum herbs, can be reasonable to guide therapy, and to provide a scientific evaluation basis for the integrated control of the quality of Liao asarum herbs...
