Objective:We have proved that methyl linoleate can effectively inhibit the expression of leukocyte inflammatory cytokines with the methord of cell culture. And on the basis of this study, we want to investigate the therapeutical effect of linoleic acid and methyl linoleate on acute and chronic inflammation. In order to study the therapeutical effect, we plan to prepare mice ear edema model and adjuvant arthritis of rat model (AA). Through this study, we hope to lay a foundation for new anti-inflammatory drug development.Method:First embrocate medicine (atoleine, dexamethasone, linoleic acid and methyl linoleate) on the right ears of the mice, and3days later, use croton oil to prepare the mouse ear edema model. During this period, observe general conditions of the mice. In the end, cut off the double ears and weigh the right ear and the left ear. Then compare the swelling degree of different groups, and calculate the inhibition of swelling degree. In the same time, observe the concentration of IL-β,TNF-α and NO in the mice’s ears. So we can know if linoleic acid and methyl linoleate have prophylactic effect to acute inflammation.Use CompleteFreund’s adjuvant (CFA) to prepare adjuvant arthritis rat model (AA), then through oral administration and embrocate medicine for drug treatment. After28days, compare the joint swelling degree, body weight, thymus index, spleen index, and the concentration of IL-1β and TNF-α in serum. Finally analyze the therapeutic effect of linoleic acid and methyl linoleate on chronic inflammation.Result:The acute anti-inflammatory results show that:the embrocating of linoleic acid and methyl linoleate on the ear can cause skin redness, dryness and thickeness, but have an effectively inhibit effect on croton oil-induced ear swelling of mice. And we also find that in the groups of linoleic acid and methyl linoleate, the concentration of IL-1β, TNF-α and NO in the ears are lower than the model group, but there is no sense in statistics. These results indicate that linoleic acid and methyl linoleate have preventive effect on acute inflammation, and methyl lionleate has weaker skin irritation.The chronic anti-inflammatory results show that:linoleic acid and methyl linoleate can obviously decrease the levels of IL-1β and TNF-α in serum, and can significantly improve arthritis symptoms in AA rats, reduce synovial inflammation. In the same time they do not have side effects, such as weight loss and inhibition of systemic immunological lead by dexamethasone. The effect of Low doses of linoleic acid and methyl linoleate will be better to chronic inflammation.Conclusion:Linoleic acid and methyl linoleate can reduce the levels of related inflammation factors, such as IL-1β, TNF-α and NO. And have obvious improvements for acute and chronic inflammation. But when topical administration, they have a stimulating effect on skin, they can cause redness, dryness, thickening. And methyl linoleate has lower stimulation effect than linoleic acid.
