Objective:To investigate the effect of ERK1/2through Pentylenetetrazole (PTZ)-kindled epilepsyon rats learning and memory and hippocampus dentate gyrus cell proliferation.Methods:Male rats of68weeks old weighted180220g are randomly divided into threegrou-ps,normal control group(intraperitoneal injection of saline,the lateral ventricleinjected saline),two model groups(intraperitoneal injection of saline group, the lateralventricle injected separately saline,U0126). Chronic modelof epilepsy was induced by1%PTZ of35mg/kg every day30consecutive days intraperitoneal injection. The standardis completely lit by Racin grading standard for3consecutive level IV or more sportsconvulsions. The three groups rat behavioral change was assessed by the Morris watermaze,observation form structure change in the hippocampus by HE dyed, Byimmunohistochemical stains observation of neurons proliferation in the hippocampus.During the experiment, continuous lateral ventricle note medicine.Results:1.Morris water maze orientation navigation experiments, After five days oftraining,each group Escape Latency was corresponding shorten, The average incubationperiod of epilepsy group (PTZ+NS)was significantly longer than the control group(p<0.01).The average incubation period of U0126group significantly extend comparing with theepilepsy group (PTZ+NS)(p<0.05),thus the number of through platform and swimmingtime of goal quadrant significantly shorten(p<0.05).2.Chronic epilepsy group of big rat existed hippocampal neurons loss andkaryopyknosis,thus U0126aggravated neurons damages futher. the hippocampus BrdUpositive cells of chronic epilepsy group is more than the control group(p<0.05). Thehippocampus Brdu positive cells of U0126group (PTZ+U0126) is less than epilepsy group (PTZ+NS)(p<0.05),the group of injecting U0126and orexin-A significantlyincrease comparing with U0126group(p<0.05).Conclusion:Epilepsy rats function of learning and memory was damaged by certain degree,Orexin-A improve epilepsy rats the function of spatial learning and memory,ERK1/2antagonist U0126accentuate further thelesion of spatial learning and memory,partlyinhabit the function of orexin-A,Orexin-A improve epilepsy rats learning and memoryfunction associated with promoting the hippocampus nerve cell proliferation concern,ERK1/2 way activation play an important role.
