A. pendulum as one commonly used ethnic medicine is the radices of A. pendulum Busch. and A. flavum Hand.-Mazz., which have effects of relieving pains and expelling wind and moist, and is widely used to treat rheumatoid arthritis wounds, etc.A. pendulum is a poisonous medicinal materials,the Qiang ethnic minority has special processing technique of sand fried method with "reducing toxicity and keeping efficacy" role. However, at present the quality standards studies of the sand fried processed products are lacking, and the "reducing toxicity and keeping efficacy’ characteristic components of intestinal flora metabolism is not clear.Objective:This paper based on the ethnic medicine A. pendulum sand fried processed products as the research object. In the establishment of its quality standard study based on, in order to clear the medicinal components and metabolism rules of sand fried processed products, using serum pharmacochemistry and in vitro intestinal flora metabolism method.Method:1. Selection of A. pendulum sand fried processed products, using thin layer chromatography, high performance liquid chromatography and determination of moisture content and methods, respectively, on its benzoyl aconine, moisture, ash, extracts and heavy metal content determination, establishment of A. pendulum sand fried processed products quality standards; at the same time, combined with the HPLC fingerprint study on the quality of processed products.2. Using HPLC-DAD/LC-MS to characterize the metabolites in serum of rats after oral administration of total alkaloid of A. pendulum sand fried processed products.3. Using in vitro intestinal flora metabolism method, combined with HPLC-DAD, study of Aconitum pendulum sand fried processed products of characteristic components of benzoyl aconine intestinal flora metabolism.Result:1. The the content of benzoylaconine in the herbs was0.053-0.173%,The average of moisture in the herbs was2.08%,and the total ash content was2.89%,The content of acid-insolubal was1.08%.The content of70%ethanol extracts was21.38%,There is no aconitine in the herbs.2.2components absorbed into the bloodstream in total alkaloid of A.pendulum sand fried processed products, one of which is16-epi-pyroaconitine, benzoyl aconine were not detected in blood.3.Under the condition of intestinal microflora in vitro, benzoylaconine occur metabolism, metabolic products need to be further studied.Conclusive:1.To ensure the clinical medication safety and effectiveness, established the quality standard of A.pendulum sand fried processed products.2. A. pendulum sand fried processed products of characteristic components of16-epi-pyroaconitine can be absorbed into the blood.3. Benzoylaconine can not be absorbed into the blood, combined with intestinal flora metabolism experimental results, confirmed its to be absorbed into the blood after metabolismed by intestinal flora.
