Pine pollen as a kind of traditional food and medicine function has a wealth of nutritional value and variety of biologically active; it has broad prospects in terms of clinical application or development of health care products. Because of the more and more widely used and weak basic research of pine pollen, the subject do the studies on the method of quality including assessment resource survey and identification^purification and identification of active ingredients、the determination of effective parts、the determination of index component,fingerprints、oxidation resistance of the biological potency evaluation in order to establish the quality evaluation system of pine pollen and provide the basis for the pine pollen quality standards. Meanwhile it can provide conditions for the development of pine pollen food, health care products, Pharmaceuticals, cosmetics and additives.Ⅰ、Identification of pine pollenThe main original plants of the pine pollen circulation on the market arePinus massoniana Lamb、Pinus tabulae formis Carr. And pinus yurmanensis Faranch. Meanwhile because of the pollen wall which can affect the chemical composition of the extraction, the price between unbroken and broken pine pollen is different.Unbroken and broken pine pollen can be distinguished by properties identification and microscopic identification. Adulteration and the long diameter of Pollen grains and the airbags and short diameter of Pollen grains can be detected by microscopic identification.Ⅱ、Studies on chemical constituents of pine pollenTwo compounds are identified after the isolation and identification of Pine pollen:Naringenin and dotriacontanoalcohol which was first identified in Pine pollen, naringenin and rutin content of pine pollen from13different areas were determined by HPLC. The result showed that there was high content of naringenin and low content of rutin in pine pollen.the average content of naringenin and rutin is0.207mg·g-1and0.0014mg·g-1.the content of naringenin of different areas had a big difference and the content of rutin has small difference. UV spectrophotometry was used to determine the total flavonoids contend total sterols、total polysaccharides. the content of the total flavonoids and total sterols of different areas had a big difference and the content of total polysaccharides has small difference.Ⅲ oxidation resistance of biological titer evaluation researchUV spectrophotometry was used to determine scavenging activity of DPPH, hydrogen peroxide of pine pollen from different areas, the extracts of pine pollen showed antioxidant activities of scavenging2different free radicals with obvious concentration-scavenging capacity relationship. The IC50of scavenging DPPH of nature and broken pine pollen were14.86g-L-1、12.95g·L-1separately, The IC50of scavenging hydrogen peroxide nature and broken pine pollen were20.31g·L-1、3.45g·L-1.Ⅳ、 Association analysis between antioxidant activity and main components of pine pollenMultivariate correlation analysis method was used to process date. The content of total flavonoids、total sterols、total polysaccharides was negatively correlated with the IC50of scavenging DPPH and hydrogen peroxide, the content of total flavonoids was maximal relevance and the content of total polysaccharides was minimal relevance.The correlation analysis showed that naringenin content of pine pollen associated larger with their anti-oxidative activity. So we suggested that the determination of naringenin was added to the pine pollen quality standards.Ⅴ、Studies on HPLC fingerprint of pine pollenThe studies on the fingerprint and cluster analysis of Pine pollen showed that similarity of pine pollen fingerprint was more than95%. The cluster analysis showed that there was difference between pine pollen of Pinus massomana Lamb and pimus yunnanensis Faranch, and small difference between unbroken and broken pine pollen. The establishment of HPLC fingerprint of pine pollen makes the quality standards more comprehensive and intuitive.
