Objective:Based on acupuncture theory, combined with molecular biology methods, to explore the electro-acupuncture adjustment of hypothalamic CRH mechanism of HPA axis in chronic fatigue rat.Methods:SD rats were randomly divided into normal control group、model group、model electro-acupuncture group、model Sham injection group、model CRH injection group、model CRH injection and electro-acupuncture group、model Antagonist-CRH injection group、 model Antagonist-CRH injection and electro-acupuncture group. Using the methods of chronic restraint and cold water swimming to make fatigue model. The following five groups with Intracerebroventricular administration of surgery separately, model electro-acupuncture、model CRH injection and electro-acupuncture group、model Antagonist-CRH injection and electro-acupuncture group with electro-acupuncture on BL23、ST36.To detect and analysis the ethology、 expression of hypothalamic CRH and content of ACT、CORT in Plasma after treatment.Results:1.exhaustive swimming time:After modeling, normal control group compared with other groups, time to extend, there were significant statistical differences(P<0.05); after treatment, model group compared with model electro-acupuncture、model CRH injection group and model CRH injection and electro-acupuncture group, time to shortened, there were significant statistical differences(P<0.05); model CRH injection group compared with model Antagonist-CRH injection group, time to extend, there were significant statistical differences(P<0.05); model CRH injection and electro-acupuncture group compared with model Antagonist-CRH injection and electro-acupuncture group, time to extend,there were significant statistical differences(P<0.05);2. Morris water maze test:Finding the platform time:After modeling, normal control group compared with other groups, time to shortened.there were significant statistical differences(P<0.05); after treatment, model electro-acupuncture group and model CRH injection and electro-acupuncture group compared with model group, time to extend, there were significant statistical differences(P<0.05);3. Content of ACTH in Plasma:after treatment, model group、model Sham injection group、model CRH injection group、model CRH injection and electro-acupuncture group、model Antagonist-CRH injection group、model Antagonist-CRH injection and electro-acupuncture group compared with normal control group, Content of ACTH in Plasma is decreased, there were significant statistical differences(P<0.05);4.Content of CORT in Plasma:after treatment, model group、model Sham injection group and model Antagonist-CRH injection group compared with normal control group, Content of CORT in Plasma is decreased, there were significant statistical differences(P<0.05);5. The gray values of hypothalamic’s CRH:after treatment, model group、model CRH injection group and model CRH injection and electro-acupuncture group compared with model group, The gray values of hypothalamic’s CRH is decreased there were significant statistical differences(P<0.05); model CRH injection group and model CRH injection and electro-acupuncture group compared with model electro-acupuncture group, The gray values of hypothalamic’s CRH is decreased, there were significant statistical differences(P<0.05); model electro-acupuncture group、model CRH injection group、 model CRH injection and electro-acupuncture group compared with model Sham injection group、 The gray values of hypothalamic’s CRH is decreased there were significant statistical differences(P<0.05); model Antagonist-CRH injection group and model Antagonist-CRH injection and electro-acupuncture group、model CRH injection and electro-acupuncture group compared with model CRH injection group, there were significant statistical differences(P<0.05);Conclusions:Chronic restraint and cold water swimming has been made rats body and psychological fatigue, chronic model was successful. Rats with chronic fatigue existence of abnormal HPA axis, showing reduced expression of hypothalamic CRH, content of ACTH、CORT in Plasma decreased; The rat’s body and psychological fatigue was improved after CRH expression increased, After electro-acupuncture treatment and CRH activation of the HPA axis, the rat’s body and psychological fatigue was improved; Hypothalamic’s CRH expression levels increasing, the trend of content of ACTH and CORT in plasma increasing, and chronic fatigue rat HPA axis abnormalities be some recovery.
