| Objective:To study the relationship between Apolipoprotein E gene and TOMM40gene polymorphism with longevity in Hainan centenarians.Methods:1.There were determined APOE gene polymorphism by multiplex polymerase chain reaction and gene sequencing technology in259subjets of case group and529subjets of control group.2.There were determined TOMM40gene polymorphism by polymerase chain reaction in259subjets of case group and529subjects of control group.Results:1.By comparison in two groups,APOEε2ε3,ε2ε4genotypes frequency in centenarian group were lower than in control group,which were statistically significant(P<0.05).APOE ε3ε3,ε2ε2,ε3ε4,ε4ε4genotypes were no statistically significant in two groups(P>0.05).ε2allele frequency in control group was higher than in centenarian group, which was significant difference (P<0.01);ε3,ε4alleles frequency distribution in two groups were no statistically significant (P>0.05).By gender stratification analysis,in female APOEε2ε2,ε3ε4genotypes frequency in centenarian group were higher than in control group,which were statistically significant (P<0.05);APOEε2ε3,ε2ε3,ε2ε4, ε4ε4genotypes were statistically significant in two groups (P>0.05).APOEsε3ε3genotype frequency in male centenarians group was higher than in control group, which was statistically significant (P<0.05);APOEε2ε3,ε2ε4genotypes frequency in control group were higher than in centenarian group,which were statistically significant (P<0.05);ε2allele frequency in control group was higher in centenarian group group, which was significant difference (P<0.01). By national stratified analysis,in Han nationality APOEε3ε3genotype frequency in centenarian group was higher than in control group,which was statistically significant(P<0.05); APOEε2ε2, ε2ε3,ε2ε4genotypes frequency in control group were higher than in centenarian group,which were statistical significance (P<0.05).ε2allele frequency in control group was higher than in centenarian group,which was statistically significant (P<0.05).In Li nationality APOEε3ε4genotype frequency in centenarian group was higher than in control group,which was statistically significant (P<0.01).2.By comparison in two groups,TOMM40genotypes frequency distribution were statistically significant (P<0.05),alleles frequency distribution were no statistically significant (P>0.05).By gender stratification analysis,in femele GG,GA,AA genotypes frequency was not statistically significant in comparison of two groups (P>0.05).In male AA genotype frequency in the centenarian group was higher than in control group,which was statistically significant (P<0.05). In male A allele frequency in centenarian group was higher than in control group,which was statistically significant (P<0.05). By national stratified analysis,in Han nationalityGA,AA genotypes frequency in centenarian group were higher than in control group,which were statistically significant (P<0.05). Allele A frequency in control group was higher than in centenarian group,which was statistically significant (P<0.01). In Li nationality GA genotype frequency in centenarian group was higher than in control group,which was significant.difference (P<0.01). According to the APOE s4and no-s4allele stratified analysis,AA genotype in APOEε4genotype groups frequency in centenarian group was higher than in control group,which was significant (P<0.01).allele A in APOEs4genotype groups frequency in centenarian group was higher than in control group,which was significant (P<0.05).GA,AA genotypes in no-APOEε4genotype groups frequency in control group were higher than in centenarian group,which were significant (P<0.05).allele A in APOEs4genotype groups frequency in control group was higher than in centenarian group,which was significant (P<0.05).Conclusion: 1.There are correlation APOEε2ε2, ε3ε4genotypes with longevity in Hainan female centenarians.ε3ε3genotype is linked with Hainan male centenarian.ε3ε3genotype is linked with longevity in Han nationality. ε3ε4genotype is linked with longevity in Li nationality.2.There are correlation between TOMM40rs207650AA genotype and longevity in Hainan s male centenarians.A allele is linked with Hainan’s male centenarians. GA,AA genotypes are linked with longevity in Han nationality of Hainan.GA genotype is linked with longevity in Li nationality.3.APOE gene and TOMM40gene linkage analysis showed that TOMM40gene rs2075650AA genotype and APOEε4are linked with longevity. Allele A and APOEε4are linked with longevity. |
