Study On Preventive And Therapeutic Effect Of Probiotic Lactobacillus Casei Zhang On Type2Diabetes In Rats

With a sharp increase in prevalence all over the world, diabetes has been the third killerchornic non-communicable disease threaten to human health followed after cancer andcardiovascular disease. Worsely, Type2diabetes dominate about90%of all diabetes.Nowadays, stragtegies of prevention and therpy for Type2diabetes focus on life styleconsultation and using drug for adjuvant therapy. In the present paper, preventive andalimerative effect of probiotic L. casei Zhang on T2DM rats was investigated. In addition,influence of L. casei Zhang on electrolyte of rat plasma and urine was also studied to explorenovel approach for prevention and therpy of Type2diabetes.8-week old male Sprague-Dawley rats were involved and randomly divided into fourgroups including control group, model group, preventive and therapeutic group. In this study,T2DM model was induced by2-week high fat diet plus with low dose of STZ injection. L.casei Zhang administration with a dose of4109cfu/d·rat started before high fat diet intakein preventive group and after STZ induced model fomation in therapeutic group. Duringexperimental period, physical activity, body weight, fasting and postprandial2h bloodglucose levels were monitored as well as oral glucose tolerance at model fomation and finallykilled point. Area Under Cerve was calculated from oral glucose tolerance curve. In the end,Sodium, Chloride, Potassium in plamsa and urine pH, Sodium, Chloride, Potassium, Calcium,Magnesium, Phosphorus, Ammonium, urea protein, urea creatinine, urea nitrogen and urineacid were determined.Favorable preventive effect but not therapeutic effect was observed in T2DM ratspretreated with L. casei Zhang considering fasting and postprandial2h blood glucose levels(p<0.01and p>0.05). OGTT test showed that this capacity was significantly downregulatedafter STZ injection compared to normal group (p<0.01) and significant improved OGTT wasobserved in preventive group compared to model group(p<0.01). Plasma ions test showedthat model group rats were of significantly reduced K+(p<0.01), increased Na+(p<0.01) andno change of Cl-(p>0.05), while these ions level in preventive group was of no significancein comparison to normal group (p>0.05). Urine pH in model and therapeutic group wassignificantly reduced (p<0.01) compared to normal group while in preventive group had nochange. Urine Cl-level was significantly lower in preventive and therapeutic group(p<0.01).Urine Na+, K+, Ca2+and NH4+loss was significantly improved in preventive group comparedto model group (p<0.05, p<0.01, p<0.01and p<0.01, respectively), while urine Mg2+andPhosphorus levels were of no significant changes in preventive group compared to modelgroup (p>0.05). Urea protein, urea creatinine, urea nitrogen and urine acid levels had nochange among four groups(p>0.05). It can concluded from these results that L. casei Zhang supplement have a positive effecton electrolyte disturbance of plasma and urine in T2DM rats suggesting that it is an effectiveway for diabetes prevention.
