| Objective:To observe the airway inflammation and mucus secretion in asthmatic mice.Methods:Twenty clean BALB/c mice were randomly divided into two groups:asthmatic model group (AS group n=10), Saline control group (NS group,n=10). asthmatic model group was sensitized and challenged with ovalbumin (OVA), and the Saline control group was sensitized and challenged with normal sodium, The other dealing with the asthmatic group. Total cells and differential inflammatory cells were counted in bronchoalveolar lavage fluid (BALF). The levels of IL-5and IL-32in BALF were determined by ELISA. The pathomorphological Changes in the airway and lung were observed by hematoxylin-eosin staining(HE). The goblet cell of airway wall and mucus secretion were observed by alcian blue-periodic acid Schiff staining(AB-PAS). The expression of Mucin-5AC(MUC5AC) in the airway and lung was observed by immunohistochemical staining(IHC). The expressions of MUC5AC mRNA and protein in the airway and lung were observed respectively by realtime fluorescence quantitative reverse transcription polymerase Chain Reaction (RT-PCR) and western blotting.Results:The total cells, neutrophile granulocyte, lymphocyte, and eosinophil cells in BALF of AS group were higher than those of NS group(P<0.05).The levels of IL-5, IL-32in BALF, AB-PAS-positive staining area and the expression of MUC5AC mRNA and protein in lung tissue were higher than those of NS group (P<0.05).Conclusion:Airway inflammation and mucus hypersecretion are present in asthmatic mice. The pathomorphological Changes in the airway and lung were observed by hematoxylin-eosin staining(HE). Compared with ET group, the difference was not statistically significant (P>0.05); the defferences between SIM and DEX groups were not significant(P>0.05).Conclusion:Simvastatin could inhibit the inflammation in airway of asthmatic mice.And it also reduced the secretion of mucus in airway. |
