| Objective:To explore the influence on gastric mucosa sIgA and serum IgG in newborn BALB/c mice after oral inactivated H. pylori vaccine.Methods:Fifty-six BALB/c mice were randomly divided into four groups, including PBS control group (group I), the group of inactivated H.pylori (group â…¡), the group of inactivated H.pylori and Escherichia cole heat-labile enterotoxin B subunit (LTB)(group III), and normal group (group IV). Newborn BLAB/c mice within24hours were immunized through oral gavage with corresponding antigen, Two weeks after final immunization, eight immunized mice per group were randomly selected to analyze sera IgG and gastric moucosal slgA tilters using an enzyme-linked immunesoubent assay(ELISA). Simultaneously, the remaining mice were challenged with1×108cfu live H.pylori strain SSI, twice once day, for2days. The twenty-eighth day post-challenge, all of mice that remaining were sacrificed and stomachs were removed for grades of inflammation and The counting of H.pylori colonizing.Results:The mean values of Serum IgG tilter levels were2.1±0.67ã€8.84±1.07ã€37.50±4.87ng/ml in group â… , group â…¡ and group â…¢, respectively, Thus, serum IgG tilters of group III was much higher than the other two groups, and the results showed a statistically significant difference (P<0.05). the mean values of mucosal sIgA tilters of these groups were1.60±0.49ã€1.96±0.47ã€6.56±1.24ng/ml, respectively, there was no statistically significant difference for mucosal sIgA tilters among these three groups (P>0.05). The mean scores of stomach tissue inflammation of group â… , group statistically significant difference as well as group IV were2.25±0.46ã€2.12±0.35ã€1.5±0.53ã€0.25±0.46, respectively, and the score of inflammatory activity of the above four groups were2.12±0.35ã€2.12±0.35ã€1.37±0.52ã€0.25±0.46. Among these four groups, the results were statistically significant between any two groups except the result between group â… and group â…¡. Finally, H.pylori colonization quantity in the stomachs of group â…¢ was lower than other group â… and group â…¡ (P<0.05), as expected, the H.pylori colonization was undetectable in group â…£.Conclusion:1. It is can induce special IgG and sIgA significantly increased after oral gavage with inactived H.pylori plus LTB to newborn mice.2. The that oral gavage with inactived H.pylori plus LTB to newborn mice may be associate with the lightening of inflammation and the decrease of H.pylori colonizing number. |
