ObjectiveTo observe the effects of salvia miltiorrhiza Bge on the kidney p38MAPKsignal channel and down stream signals’ change induced by intestinalischemia/reperfusion in rats,and to investigate SMB of probably effectivemechanism,in order to search for clinical new target of intestinalischemia/reperfusion treatment,and provide a theoretical basis.MethodsNighty-six male Wistar rats were randomly divided into4equal groups:sham operation group, ischemia reperfusion group, SMB group,and ischemiapostconditioning group.Intestinal ischemia/reperfusion model was constructedby incarcerating rat SMA. Enzyme-linked immunosorbent assay (ELISA) wasused to determine protein content of TNF-α and IL-1β in kidney tissue.Western blot was used to determine the expression of P-p38MAPKs.Ultraviolet spectrophotometry was used to determine the activity of plasmaDAO. HE staining morphology method was used to evaluate injury severity forintestine and kidney. The biochemical analyser was used to test BUN and Crat the four time point1,3,6,12hour of reperfusion. ResultsIn the aspect of plasma BUN,Cr and DAO’s expression with the timechanging,IRI group have a gradual increase to the3h’s top point and thengradually decreased until12h,but the index values in12h are still higher thanSham group(every point s are all have P<0.01).SMB group,compared with IRIgroup,have a significant cut-down(P<0.05).While compared with IPOgroup,every indexes are all more decreased in every time point.ELLSAmethod test kidney’s TNF-α、 IL-1β with the same tendency of aboveindexes.The kidney’s p38MAPKs active expression,selecting the mostsignificant change’s time point-3h as the study object,showed that IRI grouphave a significant increase compared with Sham group(P<0.01),SMB groupand IPO group are decreased compared with IRI group,and the change havestatistical significance(P<0.05).Getting study further to quantitive investigatethe relativity of TNF-α,IL-1β and Phospho-p38MAPKs,results showed thatPhospho-p38MAPKs and TNF-α have a positive relativity(r=0.841,P<0.01),Phospho-p38MAPKs and IL-1β have a positive relativity(r=0.898,P<0.01),and TNF-α and IL-1β also have positive relativity(r=0.872,P<0.01).HE staining showed that the pathological and ultra-structurechanges of kidney in IRI group were more significantly severe than Shamgroup and SMB and IPO group are sligher than IRI group,the SMB grouphave a lowest injure.Immuno histochemistry showed that Sham group havefew expression in kidney.IRI group have a significant expression ofPhospho-p38MAPKs and SMB and IPO group have a lower expressioncompared with IRI group.ConclusionsIRI produces the bacteria anatoxine shift, a lot of oxygen radicals, cellgene and inflammatory media as the bowel blood for the recovery of his cycleinto the kidney, then activated the p38MAPKs signal, promoted TNF-α、IL-1β cytokine release in organs and organizations for the damage, and theinflammation. Salvia Miltiorrhiza Bge can relieve kidney injury by intestinalischemia/reperfusion through the method of decreasing TNF-α、IL-1β. Themechanism is possibly connected with the inhibition effect on the kidneyp38MAPK signal channel to reduce the release of TNF-α and IL-1β.SalviaMiltiorrhiza Bge may have a better effect than ischemia postconditioning.
