By Liquid-liquid Extraction, silica gel column chromatography, TLC,HPLC, six compounds were isolated from Panax japonicus C.A.Mey. planted inHubei province. On the basis of the chemical properties and spectral data theywere identified as leanolic acid-28-O-β-D-glucopyranoside (1),3-O-(6’-butyl)-β-D-glucopyranoside aldehyde-oleanolic acid-28-O-beta-D-glucopyranoside(2), Chikusetsusaponin Ⅴ (3), Chikusetsusaponin Ib (4), ChikusetsusaponinⅣ (5) and Chikusetsusaponin Ⅳa (6). Compounds2was obtained fromPanax japonicus C.A.Mey. for the first time, and compound1was isolated forthe first time from Panax japonicus C.A.Mey. planted in Hubei province.HPLC-DAD method was developed to establish the fingerprint of Panaxjaponicus C.A.Mey., and15samples from various batches were analyzed.Furthermore; similarity calculation software, the principal component analysisand fuzzy poly-class method were used to differentiate and evaluate the wholeand partial fingerprints. The results showed that the method was accurate,reliable and contain vast information. The similarity of chromatographyfingerprints of various batches was0.848~0.991, as there are some connectionand difference between them. By matching chromatographic pattern andretention time correction, the characteristic mode of fingerprint of Panaxjaponicus C.A.Mey. was established, and the chromatography fingerprintssimilarity between various batches and characteristic mode was0.902~0.991, as this reference fingerprint can be used as quality control standardsfingerprints. And by comparing with standards, eight peaks were identified tofurther enhance the credibility of the results. Based the model ofcomprehensive assessment, the quality of Panax japonicus C.A.Mey. fromRongshui Guangxi was the best, and then the15batches of samples wereclassified into3cluster by cluster analysis.The content of Chikusetsusaponin Ⅴ, Chikusetsusaponin Ib,Chikusetsusaponin Ⅳ and Chikusetsusaponin Ⅳa in various batches weredetermined and analyzed by HPLC. The results showed that the content ofChikusetsusaponin Ⅴ was3%~7.9%, Chikusetsusaponin Ⅳ was0.2%~8.6%,Chikusetsusaponin Ⅳa was0.5%~6.2%, and Chikusetsusaponin Ib in15batches had bigger content difference.
