| Due to the vehicle’s general, construction industry development, the increase in the number of elderly, bone damage on the influence of human is becoming more and more serious, leading to bone damage seriously affected people’s quality of life. The main components of natural bone are hydroxyapatite and collagen. Marine collagen has many good characteristics. Such as, it dissolves instantly in acid and alkali solution, low immunogenicity and good thermal stability. Hydroxyapatite as artificial bone material have natural advantages, it can satisfy the biological activity and biocompatibility of implants. This study is to, collagen and hydroxy apatite as the main raw material, prepare composite bone repair materials and study its feasibility.1To the frozen shark’s skin, we measure hydroxy proline and calculate the content of collagen. In shark skin, collagen quality percentage is about9.21%. Among, the pepsin soluble collagen extraction is efficiency higher than acid soluble collagen. Utilizing SDS-PAGE electrophoresis, the largest protein peptide of the shark skin collagen is about110KDa, two smaller molecular are about80KDa and75KDa. Utilizing DSC measures the collagen denaturation temperature, and two kinds of collagen have the same denaturation temperature, at32℃.2A3-factor,3-level Box-Behnken experimental design was taken, and the response surface analysis was applied to determine the effect of casual factors on the yield of collagen from shark skin. Experiments were randomized to maximize the effects of unexplained variability in the observed responses, due to extraneous factors. The optimal conditions were as follows:the pepsin content (w/v) was1.07%, the acetic acid concentration was1.12M, the extracting time was44.17h, and the yield of collagen was9.45%. 3Using the single factor test, we can determine the optimal conditions of the electrostatic spinning. The better conditions are:collagen content10%, voltage12kV, injection rate1.0ml/h. In order to find more accurate optimal conditions, we used the orthogonal test, L9(34) orthogonal test. According to the test, we get the test results, which are the SEM photographs of spinning electron. The optimal spinning result is10.0, and we observed the spinning result of the different conditions and gave the corresponding numerical. The treatment group of spinning numerical is a lot of subjectivity, this also is the shortcoming. Through calculation and analysis, we got the influence order:collagen protein content> voltage> injection rate. The optimal spinning conditions are:collagen content11%, voltage11kV, injection rate1.1mL/h.4With collagen nanfiber support, a large amount of hydroxyapatite deposit, form chemical bonds and get the composite material, which are relatively stable and has certain characteristics. The composite was detected, using scanning electron microscopy, infrared spectrometry, X-ray diffraction, mechanical properties and cell compatibility. The results show that, the diameter of nanfiber material is unifomi thickness, has fine mechanical properties and good cell compatibility. Based on the materials’ preliminary study, we can determine that they have great prospects in the bone repair wounds. |
