Object:To screen the prescription of arginine ibuprofen cream. To establish the method of its determination and related substances. At the same time, investigation the stability of cream.Methods:Prescription are grading-up screened with L9(34) orthogonal design table.We select three influencing factors such as azone, propylene glycol and sodium laurylsulfate, the level of azone1%,3%,5%and the level of propylene glycol5%,10%,15%and the level of sodium laurylsulfate0.5%,1%,1.5%. We use Frans disperser and skin of abdomen with mice, T=37±1℃,rotation speed500r/min,determine the permeate rate, and compare with ibuprofen cream. With HPLC,a Diamonsil-C18column was used with the mobile phase of methanol-buffer phosphate(adjust pH=3.0with H3PO4),at the detection wavelength of220nm. According to2005Pharmacopeia, investigation the stability of cream.Results:Optimized prescription’ permeate rate is better than else,the standard curve is Q=472.90t-77.548and the correlation coefficient is0.9991. In determination of cream,the standard curve is C=5×10-5A-0.2569and the correlation coefficient is0.9997, area of arginine ibuprofen have a good linearity with concentration in the rang of10-80μg/ml.The contents of related substances in cream are all less than1%.The determination and related substances are standard.According to the experimental result of stability, the cream is stable in temperature and illumination.Conclusions:The formula is reasonable, prescription technology is approved. We can conclude the zero level equation from the Q-t diagram of curves. The proposed method is well highly sensitive,well accurate and simple and well stability.
